Summary: Efficient translation of human induced pluripotent stem cells (hiPSCs) into scalable cell manufacturing strategies was achieved through the establishment of a robotic platform. Automated essential steps of hiPSC culture and differentiation were conducted under chemically defined conditions. The approach enabled rapid and standardized production of billions of hiPSCs from various patient-and disease-specific cell lines, as well as automated multi-lineage differentiation into functional neurons, cardiomyocytes, and hepatocytes. Comprehensive molecular and cellular characterizations were performed to validate the robotic cell culture operations and benchmark industrial-scale cell culture for disease modeling, drug screening, and cell therapy.

COVID-19 pandemic and worldwide organ transplantation: a population-based study

Summary: The study evaluated the impact of the COVID-19 pandemic on worldwide organ transplantation activities and found an overall decrease in transplant activity in all countries studied. Kidney transplantation was most affected, followed by lung, liver, and heart transplants. There was a marked reduction in transplant activity during the first 3 months of the pandemic, with losses stabilizing after June 2020 but decreasing again from October to December 2020.

Organ donation and transplantation: a multi-stakeholder call to action

Summary: This study established a platform utilizing a humanoid robotic system to conduct iPSC experiments, successfully automating multiple processes with excellent reproducibility and flexibility. The robot system showed high accuracy and robustness, able to maintain undifferentiated iPSCs for an extended period and achieve a high differentiation rate with muscular disease patient-derived iPSCs. The expandability and flexibility of the robot system allowed for experimentation with multiple cell lines simultaneously.

Summary: The study evaluated the therapeutic potential of induced pluripotent stem cell (iPSC)-derived photoreceptor precursors in non-human primates. The results demonstrated good biocompatibility in naive models and the ability of the cells to survive and mature into cone photoreceptors in the damaged models. Optical coherence tomography showed restoration of retinal ellipsoid zone post-transplantation, indicating the safety and therapeutic potential of these clinically compliant iPSC-derived cells.

Transplanted pluripotent stem cell-derived photoreceptor precursors elicit conventional and unusual light responses in mice with advanced retinal degeneration

Summary: The study demonstrates the ability of pluripotent stem cell-derived cone precursors to engraft and restore light responses in a mouse model of end-stage photoreceptor degeneration. While half of the transplanted mice exhibited improved visual behavior, some retinal ganglion cells showed unusual responses, indicating the need for further research to improve engraftment yield and counteract retinal remodeling for clinical applications.
