In vitro static and dynamic cell culture study of novel bone scaffolds based on 3D-printed PLA and cell-laden alginate hydrogel

Summary: The aim of this study was to design and fabricate a novel composite scaffold using 3D-printed polylactic acid-based triply periodic minimal surfaces (TPMSs) and cell-laden alginate hydrogel. The scaffold improved the mechanical properties and pore size suitability for bone regeneration applications. The study used an implicit function and fused deposition modeling to generate and print the TPMS scaffolds. Micro computed tomography was employed to assess the microstructure, while compression tests were conducted to investigate the mechanical properties. The study also examined the biological response in terms of cell viability, proliferation, and attachment, and compared the results to scaffolds without alginate.

The main pro-fibrotic pathway involved in cardiac fibrosis, TGF-beta 1, may cooperate with the Hippo transcriptional pathway through YAP activation. By developing a 3D model of cardiac extracellular matrix remodeling, researchers demonstrated that inhibiting the YAP complex can reverse the pro-compaction phenotype and matrix remodeling activity induced by TGF-beta 1.
