Regioselective reductive transamination of peptidic amides enabled by a dual Zr(IV)-H catalysis

Existing catalytic modes for amide transformations include metal-catalyzed carbon-nitrogen bond activation, transamidation, and catalytic amide reduction. This study presents a novel catalytic protocol for the reductive transamination of amides to amines of exchanged identities, involving the intermediacy of aminals. The method enables the synthesis of various mono- and diamines with good yields, high functional group compatibility, and chiral retention.
