Cysteine-Activated Small-Molecule H2Se Donors Inspired by Synthetic H2S Donors

Summary: The importance of selenium in biology and health is becoming increasingly clear. This study demonstrates that selenocyclopropenones and selenoamides can release hydrogen selenide (H2Se) upon reaction with cysteine, and the rate of release can be tuned through structural modifications. The researchers developed a quantitative assay for direct H2Se trapping and quantification. Additionally, they found that arylselenoamide can release H2Se in cellular environments. Mechanistic investigations and density functional theory calculations provide insight into the possible pathways of Cys-activated H2Se release. The well-defined chemistries of Cys-activated H2Se donor motifs have potential for studying selenium biology and developing new H2Se donors and bioconjugate techniques.

Sulfane Decreases the Nucleophilic Reactivity of Zinc Thiolates: Implications for Biological Reactive Sulfur Species

Summary: A zinc dithiolate complex supported by a [N3S2] ligand was studied as a model for zinc-mediated thiolate-disulfide exchange. The results show that there is interaction between the zinc center and the disulfide moiety, leading to activation of the S-S bond for subsequent reactions. Comparison with a previously prepared zinc tetrasulfanido complex suggests that sulfane sulfur (S-0) acts as an efficient thiolate trapping agent.

Biosynthesis of selenium-containing small molecules in diverse microorganisms

The reactions of this pathway have been elucidated, and two novel selenium-carbon bond-forming enzymes and a biosynthetic pathway for producing a selenosugar have been uncovered.

Biomimetic selenocystine based dynamic combinatorial chemistry for thiol-disulfide exchange
