Lung fibrosis is induced in ADAR2 overexpressing mice via HuR-induced CTGF signaling

Ramani Soundararajan et al.

Summary: The study reveals the important role of ADAR2 in the lungs, with overexpression leading to spontaneous lung fibrosis via HuR-mediated CTGF signaling pathway. The findings contribute to a better understanding of the function of ADAR2 in the lungs and provide potential therapeutic strategies for lung fibrosis.

A-to-I RNA editing of Filamin A regulates cellular adhesion, migration and mechanical properties

Mamta Jain et al.

Summary: This study investigates the effects of ADAR2-mediated A-to-I RNA editing of Flna on actin crosslinking, cell mechanics, cellular adhesion, and cell migration. The edited version of FLNA increases cellular stiffness and adhesion but impairs cell migration. In vitro experiments show that edited FLNA leads to increased actin crosslinking, forming actin gels with higher stress resistance, thus affecting the mechanical property and mechanotransduction of cells.

Evolutionary driving forces of A-to-I editing in metazoans

Yuange Duan et al.

Summary: This article reviews the current knowledge of A-to-I RNA editing in metazoan transcriptomes, focusing on the possible evolutionary driving forces behind the editing events. It covers the evolution of ADAR gene family, recent advances in characterizing editomes of various metazoan species, factors contributing to interspecies differences in editomes, diversifying and restorative effects of editing events on protein sequences, and possible convergent evolution of RNA editing in distantly related clades.

Interplay between A-to-I Editing and Splicing of RNA: A Potential Point of Application for Cancer Therapy

Anton O. Goncharov et al.

Summary: Adenosine-to-inosine RNA editing is a widespread post-transcriptional modification system catalyzed by ADAR enzymes, which mainly occurs in dsRNA before splicing. This review focuses on the interconnections between RNA editing and splicing in human cancer. Dysregulated RNA splicing in cancer can lead to the release of excess dsRNA into the cytosol, triggering antiviral-like responses and potentially inhibiting tumor growth. Inhibiting ADAR may enhance the effects of spliceosome inhibitors and could be a promising approach in cancer therapies targeting RNA splicing.

Landscape of adenosine-to-inosine RNA recoding across human tissues

Orshay Gabay et al.

Summary: Gabay et al. presented a highly accurate atlas of A-to-I RNA editing in humans, covering editing sites across different tissues and cell subpopulations. Most edited sites are evolutionarily conserved in non-primate mammals, indicating adaptation. Editing by adenosine deaminases alters mRNA encoded information, affecting protein-coding sequences and diversifying the proteome.

Human TKTL1 implies greater neurogenesis in frontal neocortex of modern humans than Neanderthals

Anneline Pinson et al.

Summary: This study found that the modern human variant hTKTL1, but not the Neanderthal variant, can increase the abundance of bRG without affecting bIPs, thereby affecting the generation of neocortical neurons. This indicates differences in neocortical neurogenesis between modern humans and Neanderthals.

DeepLC can predict retention times for peptides that carry as-yet unseen modifications

Robbin Bouwmeester et al.

Summary: DeepLC is a deep learning-based peptide retention time predictor that can accurately predict retention times for unmodified peptides and peptides with previously unseen modifications, addressing peptide identification ambiguity in complex mass spectrometry workflows. By using atomic composition-based peptide encoding, DeepLC is able to accurately predict retention times for a wide range of modifications, including those not seen during training, potentially enabling incorrect identifications to be flagged in proteome data analysis.

A single-cell type transcriptomics map of human tissues

Max Karlsson et al.

Summary: Advances in molecular profiling have enabled the creation of high-resolution single-cell type maps of human tissues, combining single-cell transcriptomics analysis with spatial antibody-based protein profiling. This technology allows researchers to explore the expression of human protein-coding genes and determine the number of elevated genes in different cell types, facilitating comparisons with bulk transcriptomics data. The analysis reveals distinct expression clusters corresponding to cell types with similar functions within and between organs.
