Modeling the heterogeneity of sodium and calcium homeostasis between cortical and hippocampal astrocytes and its impact on bioenergetics

Summary: Emerging evidence suggests that changes in sodium concentration in astrocytes are linked to excitability, bioenergetics, neurotransmitter uptake, and neurovascular coupling. A biophysical model was developed to understand the heterogeneity in sodium concentration and its effect on astrocytic Ca2+ signals and ATP consumption in different brain regions. The model predicts that cortical astrocytes have higher ATP consumption and are more susceptible to Na+ and Ca2+ overload compared to hippocampal astrocytes. Experimental measurements confirm the model's predictions.

However, intensity-based approaches are susceptible to artifacts caused by changes in fluorophore concentrations. This study introduces a new method called rapidFLIM, which combines ultra-low dead-time photodetectors with rapid electronics to overcome this drawback. The improved rapidFLIM allows for quantitative and dynamic imaging of low-emission fluorophores at an unprecedented speed and independent of changes in cell volume.
