Stages of OCP-FRP Interactions in the Regulation of Photoprotection in Cyanobacteria, Part 1: Time-Resolved Spectroscopy

Georgy V. Tsoraev et al.

Summary: Using time-resolved spectroscopy, this study demonstrates that the interaction between Fluorescence Recovery Protein (FRP) and photoactivated Orange Carotenoid Protein (OCP) begins early in the photocycle. The interaction of FRP with the compact OCP state prevents the formation of the signaling state capable of interacting with the antenna. This research refines the current model of photoinduced OCP transitions and the interaction of its intermediates with FRP.

Light-Harvesting Complex II Adopts Different Quaternary Structures in Solution as Observed Using Small-Angle Scattering

Maksym Golub et al.

Summary: In this study, small-angle scattering was used to investigate the solution structures of solubilized LHCII. The results showed that the solution structures of LHCII may differ under different preparation conditions, with the LHCII-beta-DM solution revealing a stable oligomer structure that may play a crucial role in photoprotective nonphotochemical quenching.

Invisible Detergents Enable a Reliable Determination of Solution Structures of Native Photosystems by Small-Angle Neutron Scattering

M. Golub et al.

Summary: Small-angle neutron scattering (SANS) provides structural information on PSI and PSII in solution under nearly physiological conditions without the need for crystallization or temperature decrease. Proper contrast matching of the detergent belt surrounding the protein is crucial for the reliability of the solution structure, with specifically deuterated detergents showing promise in this regard. Differences in temperature and solution state compared to high-resolution structures suggest the presence of a manifold of conformational substates accessible by protein dynamics under physiological conditions.

Oligomerization processes limit photoactivation and recovery of the orange carotenoid protein

Elena A. Andreeva et al.

Summary: This study investigates the photoinduced structural changes in the orange carotenoid protein (OCP) using a combination of experimental techniques. The results suggest that oligomerization plays a role in regulating the OCP photocycle, and a numbed state is formed upon non-photoproductive excitation.

Summary: This study reports on the characteristics of Plankto-OCP, a variant of the orange carotenoid protein (OCP) involved in cyanobacterial photoprotection. Plankto-OCP shows higher photoactivation and recovery rates, stronger energy-quenching activity, and is affected by the presence of a his-tag and the functionalizing carotenoid. The structures of Plankto-OCP reveal its molecular breathing motions and potential structural changes that explain its unique properties.

Structures of a phycobilisome in light-harvesting and photoprotected states

Maria Agustina Dominguez-Martin et al.

Summary: In this study, the structures of Phycobilisome (PBS) in cyanobacteria were solved using cryogenic electron microscopy. The researchers discovered a previously unknown linker protein and revealed different conformational states of the antenna. Moreover, they elucidated the energy transfer pathways within PBS and provided insights into the control of cyanobacterial light harvesting, which has implications for bioengineering light-harvesting systems.

Insights into Solution Structures of Photosynthetic Protein Complexes from Small-Angle Scattering Methods

Maksym Golub et al.

Summary: High-resolution structures of photosynthetic pigment-protein complexes are typically determined using crystallography or cryo-electron microscopy, which have limitations in studying proteins in aqueous solution at physiological temperatures. Small-angle neutron and X-ray scattering offer a solution by providing structural information for proteins in solution at room temperature. Studies on phycocyanin and Photosystem I revealed discrepancies in their solution structures compared to crystal structures, possibly due to protein dynamics at physiological temperatures.
