Application and synthesis of thiazole ring in clinically approved drugs

Summary: The development of heterocyclic derivatives has led to the production of many new synthetic and natural compounds. Thiazole, a unique five-membered heterocyclic motif, is widely used as a core skeleton in various pharmaceutically important compounds due to its diverse biological activities. Over 90 thiazole-containing derivatives are currently under clinical investigation, and some have been approved for treating diseases. Thiazole derivatives can be further explored to search for new drugs with improved therapeutic efficacy. This review provides an overview of the applications and synthetic routes of approved thiazole-containing drugs, aiming to guide the rational design of more effective drug candidates.
