Summary: We report the palladium-catalyzed aminocarbonylation of (hetero)aryl iodides using alpha-amino acid esters as nucleophiles. The synthesized N-capped alpha-amino acids are important building blocks in biology. The reaction is carried out under mild conditions, yielding products with high enantioselectivity in a short period of time. Additionally, regioselective carbonylation of histidine is achieved for the first time.

Palladium-Catalyzed Aminocarbonylation of Heteroaryl Iodides

Summary: This study reports the palladium-catalyzed aminocarbonylation of heteroaryl iodides using chloroform as a surrogate for carbon monoxide. The reaction demonstrated a broad substrate scope, successfully synthesizing quinoline-3-carboxamides and other heteroaryl amides with yields up to 90% under non-inert conditions and microwave irradiation at 80 degrees C in 30 minutes. This carbonylation method provides an efficient access to biologically important heteroaryl carboxamides that were previously difficult to obtain.
