Modification of Pyrrolo[2,1-a]isoquinolines and Pyrrolo[1,2-a]quinolines with Ammonium Acetate and Dimethyl Sulfoxide via Methylthiomethylation

Summary: An efficient methylthiomethylation of pyrroloisoquinolines and pyrroloquinolines has been achieved using ammonium acetate and dimethyl sulfoxide. Moderate to good yields of methylthiomethylated heterocycles can be obtained in most cases, with trace amounts to good yields of methylene-bridged products observed. The chemoselectivity of this process is greatly influenced by the choice of DMSO activator and its amount. It is also noteworthy that the process is scalable and the product can easily be transformed to sulfone and sulfoxide.

Summary: The synthesis of 1,4-disubstituted pyrazoles was successfully achieved through a cascade reaction between enaminones, hydrazines, and DMSO catalyzed by molecular iodine in the presence of Selectfluor. DMSO played a dual role as the C-1 source and the reaction medium, and the synthesis of 1,3,4-trisubstituted pyrazoles using aldehydes as alternative C-1 building blocks was also accomplished.

Summary: This study presents a mild and efficient method for synthesizing symmetrical and unsymmetrical diarylmethanes. The use of DMSO as a carbon source and solvent, along with the incorporation of the -CD2 moiety, allows for the synthesis of a wide range of functionalized diarylmethanes. This protocol has been successfully applied to the synthesis of an anti-breast cancer agent and an anticoagulant agent.

Summary: This method describes the unprecedented C(sp(2))-H methylation of iminoamido heterocycles as nucleoside base analogues, using trimethylsulfoxonium salt as a methylating agent under aqueous conditions. It achieves a wide substrate scope and excellent level of functional-group tolerance, and can be readily applied to site-selective methylation of azauracil nucleosides, highlighting its synthetic potential.

Formylation and Bromination of Pyrrolo[2,1-a]isoquinoline Derivatives with Bromoisobutyrate and Dimethyl Sulfoxide

Summary: An efficient formylation method for pyrroloisoquinolines using bromoisobutyrate and dimethyl sulfoxide as carbonyl reagent has been developed, providing good yields of formylated products (up to 94%) which can be scaled up easily. Additionally, the combination of bromoisobutyrate and dimethyl sulfoxide can also act as a bromination reagent for pyrroloisoquinolines without methoxy groups, yielding brominated products in acceptable to good yields (up to 82%).

Aerobic cross-dehydrogenative couplings of N-heteroarenes with toluene derivatives at room temperature

Summary: A new oxidative coupling reaction was achieved using visible-light-induced photocatalysis, providing a sustainable alternative to Minisci benzylation reactions. The combination of photocatalysis, bromo radical mediation, and aerobic oxidation proved to be essential for the success of the transformation. Mechanistic studies showed the feasibility of both oxidative and reductive quenching of the excited photocatalyst in the initial step.
