CFTR mRNAs with nonsense codons are degraded by the SMG6-mediated endonucleolytic decay pathway

Edward J. Sanderlin et al.

Summary: This study reveals that CFTR mRNAs with nonsense codons are predominantly degraded by the SMG6-mediated branch of the NMD pathway. This finding provides potential strategies for the treatment of cystic fibrosis.

Molecular profiling of individual FDA-approved clinical drugs identifies modulators of nonsense-mediated mRNA decay

Jingrong Zhao et al.

Summary: Nonsense-mediated mRNA decay (NMD) is a process that degrades transcripts with premature stop codons. It is important to study the effects of clinically approved drugs on NMD activity. This study used a sensitive method to analyze the impact of individual FDA-approved drugs on cellular NMD efficiency and found that most drugs have no significant effect on NMD but can cause transcriptional changes. Additionally, the anticancer drug HHT consistently upregulates various endogenous NMD substrates in multiple cell types.

ISRIB Blunts the Integrated Stress Response by Allosterically Antagonising the Inhibitory Effect of Phosphorylated eIF2 on eIF2B

Alisa F. Zyryanova et al.

Summary: ISRIB antagonizes the activation of the ISR by accelerating eIF2B nucleotide exchange activity, with better efficacy in the presence of eIF2(alpha P). This is achieved through antagonistic allostery in ISRIB action on eIF2B.

eIF2B conformation and assembly state regulate the integrated stress response

Michael Schoof et al.

Summary: The integrated stress response (ISR) is activated by phosphorylation of translation initiation factor eIF2 in response to various stress conditions. Phosphorylated eIF2 inhibits eIF2B, leading to ISR induction. ISRIB can inhibit ISR even in fully assembled eIF2B decamers, suggesting allosteric communication between distant binding sites.
