Ablation of Growth Hormone Receptor in GABAergic Neurons Leads to Increased Pulsatile Growth Hormone Secretion

Willian O. dos Santos et al.

Summary: This study investigated the consequences of GHR ablation in specific neuronal populations and found that GHR ablation in GABAergic neurons leads to increased GH secretion, lean body mass, and body growth. On the other hand, GHR ablation in glutamatergic neurons results in normal GH secretion but reduced lean body mass. These findings contribute to our understanding of how GH affects metabolism and are important for understanding the neuroendocrine and metabolic effects of GH.

Unexpected role for IGF-1 in starvation: Maintenance of blood glucose

Fei Fang et al.

Summary: Ghrelin is essential for the maintenance of viable blood glucose levels in starved mice, and its absence results in a reduction of plasma IGF-1. IGF-1 stimulates adipose tissue lipolysis, providing energy and substrates for hepatic gluconeogenesis in starved mice. However, this action of IGF-1 is contrary to its known inhibitory effect on adipose tissue lipolysis in fed mice. The maintenance of plasma IGF-1 in starved mice is not mediated by GH.

Summary: Growth hormone plays a crucial role in regulating metabolism in the brain, affecting food intake, energy expenditure, blood sugar levels, and neuroendocrine changes. It acts as a hormonal factor that conveys homeostatic information to the brain to produce metabolic adjustments in order to promote energy homeostasis.

Summary: Entropy has been increasingly used in various research fields to quantify the regularity, variability, and randomness of time series and image data, yet there is a lack of validated, open-source software tools. EntropyHub is an open-source toolkit that provides a wide range of functions for estimating various entropy methods, aiming to make advanced entropic time series analysis straightforward and reproducible.

Disrupting the ghrelin-growth hormone axis limits ghrelin's orexigenic but not glucoregulatory actions

Summary: The study investigated the role of acyl-ghrelin on GH secretion and metabolic actions by selectively deleting GHSR from somatotrophs. Results suggest that the direct engagement of GHSR-expressing somatotrophs is essential for acyl-ghrelin to stimulate GH secretion and influence its metabolic actions.

Towards Understanding the Direct and Indirect Actions of Growth Hormone in Controlling Hepatocyte Carbohydrate and Lipid Metabolism

Mari C. Vazquez-Borrego et al.

Summary: Growth hormone plays a critical role in regulating metabolic function by modulating the production and function of insulin-like growth factor 1 and insulin through its receptor. This hormone, along with IGF1 and insulin, coordinates metabolic control in various tissues. The review focuses on the direct and indirect actions of GH in controlling liver carbohydrate and lipid metabolism, discussing the need for further research on GH's role in metabolic health and disease.

Hypothalamic GHR-SIRT1 Axis in Fasting

Juliana Bezerra Medeiros de Lima et al.

Summary: The hypothalamus plays a key role in controlling physiological functions and whole-body metabolism. Growth hormone (GH) and the GH receptor (GHR) are expressed in hypothalamic regions involved in regulating feeding and energy balance. SIRT1, a NAD(+)-dependent protein deacetylase, interacts with GHR signaling and plays a role in sensing nutrient availability in response to caloric restriction. In the arcuate nucleus (ARC) of the hypothalamus, GHR-expressing neurons also express SIRT1 and upregulate SIRT1 expression in response to fasting, revealing a novel interaction between GH and SIRT1 in fasting responses.

ARCGHR Neurons Regulate Muscle Glucose Uptake

Juliana Bezerra Medeiros de Lima et al.

Summary: The growth hormone receptor (GHR) is expressed in brain regions involved in energy homeostasis and glucose metabolism. Activating GHR-expressing neurons in the arcuate nucleus enhances respiratory exchange ratio and promotes glucose utilization over fat, especially under fasting conditions. Increased insulin sensitivity in skeletal muscle and improved glucose tolerance are observed upon the activation of ARC(GHR+) neurons, indicating their role in regulating glycolysis and muscle insulin sensitivity in vivo.

Summary: The ablation of GHR in CRH cells does not lead to major alterations in metabolism, the hypothalamic-pituitary-adrenal axis, acute stress response, or anxiety in mice. While reduced energy expenditure was observed in female mice, the overall physiological importance of GH action on CRH neurons remains uncertain and warrants further investigation.
