(-)-Amarbellisine is none other than (-)-montanine: A CASE-3D and dereplication NMR study

Armando Navarro-Vazquez

Summary: The total synthesis of (-)-amarbellisine lycorine alkaloid in 2013 revealed significant differences in the NMR spectra compared to the originally isolated alkaloid, suggesting a different diastereoisomer structure. However, the present CASE-3D configurational analysis using DFT predictions failed to fit any possible diastereoisomeric lycorine-type structures. Reexamination of the NMR data for (-)-amarbellisine and literature search confirmed that it is actually the well-known (-)-montanine alkaloid.

ML-J-DP4: An Integrated Quantum Mechanics-Machine Learning Approach for Ultrafast NMR Structural Elucidation

Yi-Hsuan Tsai et al.

Summary: A new tool, ML-J-DP4, has been developed for efficiently and accurately determining the most likely structure of complex molecules within minutes using standard computational resources. The tool combines fast Karplus-type J calculations with NMR chemical shifts predictions enhanced with machine learning (ML) in the J-DP4 formalism. The ML predictions provided by this tool are accurate and compare favorably with other ML methods.

Real-time prediction of 1H and 13C chemical shifts with DFT accuracy using a 3D graph neural network

Yanfei Guan et al.

Summary: Nuclear magnetic resonance (NMR) is a primary technique used to elucidate the chemical structure, bonding, stereochemistry, and conformation of organic compounds. In this study, a rapid machine learning protocol was developed to predict H-1 and C-13 chemical shifts through an efficient graph neural network (GNN) using 3D structures as input, improving prediction accuracy. The model demonstrated success in various applications, showcasing its potential in enhancing organic structural assignments using NMR data.
