Triterpenoids from the leaves of Abies koreana and their biological activities

Summary: Seven undescribed triterpenoids, abikoranes A-G, along with three known triterpenoids, were isolated from the leaves of Abies koreana E. H. Wilson. The structures of these compounds were determined through various analysis methods, including NMR, HRMS, ECD, specific rotation, and DP4+. Some of the isolated compounds showed strong cytotoxic activities against cancer cells, inhibited nitric oxide production, and promoted nerve growth factor release in glioma cells.

DFT-aide d infrare d and electronic circular dichroism spectroscopic study of cyclopeptide S-PK6 and the exploration of its antitumor potential by molecular docking

Summary: In this article, the infrared (IR) and electronic circular dichroism (ECD) spectroscopic study of the isoindolinone-containing analog S-PK6 of natural antitumor cyclopeptide Phakellistatin 6 was reported. The study determined the peak positions of various amino acid residues in the IR spectrum of S-PK6 using DFT-calculations. It was found that the simulation at the TDDFT/CAM-B3LTP/cc-pVDZ level provided a decent match for the experimental ECD spectrum of cyclopeptide. Molecular docking was performed to investigate the binding mode between S-PK6 and different cancer-related proteins, revealing a target-induced conformational adaptation mechanism. This research provides critical insights into the structure of small cyclopeptides and hints towards the design of peptidyl drugs.

Forrestiacids E-K: Further [4+2]-Type Triterpene-Diterpene Hybrids as Potential ACL Inhibitors from the Vulnerable Conifer Pseudotsuga forrestii

Summary: Seven [4 + 2]-type triterpene-diterpene hybrids, named forrestiacids E-K, derived from a rearranged or a normal lanostane unit and an abietane moiety, were isolated and characterized from Pseudotsuga forrestii. Their chemical structures and configurations were determined through LC-MS/MS analysis, phytochemical procedures, and spectroscopic data. Some of the compounds showed significant inhibition against ATP-citrate lyase, indicating their potential as therapeutic agents. The findings emphasize the importance of preserving plant species diversity for the discovery of new chemical compounds and potential therapeutics.

The Sixth Mass Extinction: fact, fiction or speculation?

Summary: There have been five Mass Extinction events in the history of Earth's biodiversity, and the Sixth Mass Extinction may currently be occurring due to human activities. While some dispute the existence of the Sixth Mass Extinction, arguing that the rate of species loss is not significantly different from background rates, this viewpoint is flawed due to the bias of the IUCN Red List. Incorporating estimates of invertebrate extinctions suggests that the current rate of extinction far surpasses background rates, pointing to the start of the Sixth Mass Extinction.

Cells have evolved de novo lipogenesis (DNL) to generate fatty acids from alternative carbon sources. However, abnormal upregulation of DNL is associated with various pathologies. Inhibiting core enzymes of DNL represents a potential therapeutic strategy, and several synthetic DNL inhibitors are currently in development. Aberrant upregulation of DNL is implicated in conditions such as cardiovascular disease, metabolic disorders, and cancers.

Further research on new ACLY inhibitors is of great significance for lipid-related pathologies and cancer treatment.

Comparative transcriptomic analysis unveils the deep phylogeny and secondary metabolite evolution of 116 Camellia plants

Summary: Camellia plants have a great diversity and possess immense economic, ornamental, and cultural values. Through sequencing the transcriptomes of 116 Camellia plants, we have constructed a pan-transcriptome and resolved the phylogeny of the genus Camellia. We found that Camellia plants shared a recent whole genome duplication event and had large expansions of transcription factor families associated with stress resistance and secondary metabolism. Additionally, secondary metabolites related to tea quality were heavily accumulated in Camellia plants from section Thea.
