Autophagy-associated circRNA circATG7 facilitates autophagy and promotes pancreatic cancer progression

Zhiwei He et al.

Summary: This study investigates the role of autophagy-associated circRNA circATG7 in the progression of pancreatic cancer. The results show that circATG7 is upregulated in pancreatic cancer tissues and cells, and its expression is associated with tumor diameter and lymph node invasion. CircATG7 promotes cancer cell proliferation, mobility, and autophagy, while its knockdown has the opposite effects. Furthermore, circATG7 regulates the cancer cells' biological functions through the miR-766-5p/ATG7 pathway and the HUR/ATG7 interaction, which is dependent on autophagy flux.

Genotoxic stress-activated DNA-PK-p53 cascade and autophagy cooperatively induce ciliogenesis to maintain the DNA damage response

Ting-Yu Chen et al.

Summary: The DNA-PK-p53 cascade induces primary cilia formation under genotoxic stress, contributing to the DNA damage response, while lack of this pathway decreases genotoxic stress-induced ciliogenesis. Besides, ciliogenesis is regulated not only by DNA-PK, but also by autophagy.

Circular RNA CircMAP3K5 Acts as a MicroRNA-22-3p Sponge to Promote Resolution of Intimal Hyperplasia Via TET2-Mediated Smooth Muscle Cell Differentiation

Zhi Zeng et al.

Summary: The study demonstrates that circMAP3K5 acts as a regulator of neointimal hyperplasia by sequestering miR-22-3p to inhibit TET2 expression, and subsequently affecting vascular smooth muscle cell differentiation and proliferation. Targeting the circMAP3K5/miR-22-3p/TET2 axis may offer a potential therapeutic strategy for diseases associated with intimal hyperplasia, including restenosis and atherosclerosis.

BMP9 and BMP10 Act Directly on Vascular Smooth Muscle Cells for Generation and Maintenance of the Contractile State

Lei Wang et al.

Summary: The study revealed that BMP9 and BMP10 act directly on VSMCs for induction and maintenance of their contractile state. The effects of BMP9/10 in VSMCs are mediated by different combinations of BMP type 1 receptors in a vessel bed-specific manner, offering new opportunities to manipulate blood pressure in the pulmonary circulation.

Endonuclease G promotes autophagy by suppressing mTOR signaling and activating the DNA damage response

Wenjun Wang et al.

Summary: ENDOG is released from mitochondria during starvation and promotes autophagy by enhancing its interaction with 14-3-3 gamma through phosphorylation, leading to mTOR pathway suppression and activation of DNA damage response through its endonuclease activity.
