Structural modification aimed for improving solubility of lead compounds in early phase drug discovery

Summary: Solubility is a crucial factor in early phase drug discovery, as many potent lead compounds fail to reach clinical trials due to low bioavailability caused by insufficient solubility. Improving solubility while maintaining potency is a challenging task for medicinal chemists. Poor solubility not only hinders in vitro and in vivo assays but also prolongs drug development time and increases costs. Various techniques, including physical, chemical, and miscellaneous methods, are used to enhance solubility. The medicinal chemistry approach of structural modification is versatile and can improve other pharmacokinetic/physicochemical parameters simultaneously.

Summary: In this study, the discovery of BMS-737 as a potent small molecule inhibitor for the treatment of castration-resistant prostate cancer (CRPC) is reported. The inhibitor demonstrates high selectivity for CYP17 lyase over CYP17 hydroxylase, with a clean xenobiotic CYP profile. Extensive structure-activity relationship studies led to the identification of BMS-737, which showed significant lowering of testosterone in animal experiments.

Crystal Structure of CYP3A4 Complexed with Fluorol Identifies the Substrate Access Channel as a High-Affinity Ligand Binding Site

Summary: This study determined the crystal structure of CYP3A4 complexed with fluorol, a small fluorescent dye, and revealed the importance of the substrate channel as a high-affinity ligand binding site. Target-specific compound selectivity for multi-target drug discovery and repurposing

Summary: Most drug molecules have the ability to modulate multiple target proteins, which can lead to both therapeutic effects and unwanted side effects. Evaluating the selectivity of a compound is an important factor in drug development and repurposing efforts. Traditional methods for characterizing selectivity fall short in quantifying how selective a compound is against a particular target protein. In this study, we propose an optimization-based selectivity scoring method that allows for the identification of potent and selective compounds against given kinase targets. We demonstrate the effectiveness of this method in finding highly selective compounds in computational experiments using a large-scale kinase inhibitor dataset.

Generation of new inhibitors of selected cytochrome P450 subtypes- In silico study

Summary: Physicochemical and pharmacokinetic compound profile play a crucial role in determining the potency of a compound to become a future drug. In this study, the focus is on the metabolic stability and selected subtypes of cytochrome P450 proteins. A protocol for generating potential inhibitors of these cytochrome isoforms is developed, which involves the generation and assessment of new derivatives, docking and evaluation of compound inhibition, and the use of a graph neural network to predict activity changes. The methodology provides valuable support in the design of new cytochrome P450 inhibitors and enables instant evaluation of compound-cytochrome interactions.

Summary: This study used resonance Raman spectroscopy to compare the effect of Mn-substituted cytochrome b(5) on the oxy complex of CYP17A1, resulting in a specific allosteric interpretation for the more pronounced acceleration seen with the 17OH-PROG substrate. The presence of Mn-b led to a reorientation of a hydrogen bond between the 17OH group of the substrate and the proximal oxygen atom, favoring lyase catalysis, but this effect was not observed for 17OH-pregnenolone.

Stepwise binding of inhibitors to human cytochrome P450 17A1 and rapid kinetics of inhibition of androgen biosynthesis

Summary: In this study, the mechanisms of inhibition by drugs developed to inhibit P450 17A1 were considered, revealing a rapid and strong inhibition of both 17 alpha-hydroxylation and lyase activity upon initial binding of any of the inhibitors. The compounds bound to P450 17A1 in a multistep process, but did not show any lag in the onset of inhibition in rapid-quench experiments. These results contrast with inhibitors of P450 3A4, where complete inhibition is not observed until conformational changes are completed.

Summary: It has been known for over 50 years that cytochrome b(5) stimulates certain cytochrome P450-catalyzed oxidations, with the strongest effect seen in the P450 17A1 lyase reaction. A fluorescently labeled variant of human b5 was created to study b5-P450 interactions, revealing that the dynamics of the P450 17A1:b5 complex play a crucial role in catalytic activity. The addition of NADPH-P450 reductase to the complex resulted in a ternary P450:b5:POR complex, highlighting the importance of this interaction for optimal catalytic activity.

New benzothieno[2,3-c]pyridines as non-steroidal CYP17 inhibitors: design, synthesis, anticancer screening, apoptosis induction, and in silico ADME profile studies

Summary: This study synthesized a series of benzothieno[2,3-c]pyridine derivatives and found that one compound, 5c, exhibited potent anti-cancer activity by inhibiting CYP17 enzyme activity and regulating testosterone production mechanism to suppress prostate cancer growth. In addition, compound 5c also showed promising results in terms of cytotoxicity, cell cycle regulation, and apoptosis mechanisms. Furthermore, the ADME profile of compound 5c demonstrated good oral bioavailability and metabolic stability.
