Impact of structural dynamics on biological functions of flaviviruses

Karin Stiasny et al.

Summary: Flaviviruses, transmitted by mosquitoes or ticks, are important human pathogens. Research has shown that these viruses use a "breathing" mechanism to enhance their adaptability and replication through interactions with the immune system and host factors, and a novel mechanism of antibody-mediated infection enhancement has been identified.

Alimentary Infections by Tick-Borne Encephalitis Virus

Martina Lickova et al.

Summary: Tick-borne encephalitis virus can be transmitted to humans through ticks and the alimentary route, which involves consuming raw milk products from infected ruminants. The alimentary route of TBEV infection is recognized as a contributing factor to the increase in TBE incidence in Europe.

T-Cell Immunoglobulin and Mucin Domain 1 (TIM-1) Is a Functional Entry Factor for Tick-Borne Encephalitis Virus

Xiaowei Zhang et al.

Summary: This study identified T-cell immunoglobulin and mucin domain 1 (TIM-1) as a cellular entry factor for tick-borne encephalitis virus (TBEV), mediating the uptake of TBEV into cells. TIM-1 deficiency significantly reduced viral burden and pathogenesis, highlighting its importance in TBEV infection. This discovery provides a potential target for antiviral intervention and improves understanding of virus infection.

Molecular Determinants of Flavivirus Virion Assembly

Trisha R. Barnard et al.

Summary: Virion assembly is a crucial step in the lifecycle of all viruses, but remains one of the least understood processes for Flavivirus genus. Recent studies show that besides viral structural proteins, nonstructural proteins also play key roles in coordinating assembly and packaging of viral genome. This review highlights recent advances in understanding flavivirus virion assembly, including molecular interactions between viral structural and nonstructural proteins, host factors, and features of viral genomic RNA necessary for efficient assembly.

Pdcd10-Stk24/25 complex controls kidney water reabsorption by regulating Aqp2 membrane targeting

Rui Wang et al.

Summary: The PDCD10-STK-ERM signaling pathway was identified as a potentially novel pathway for water balance control in the kidneys, by regulating vesicle trafficking and protein abundance of AQP2. Deficiency of Pdcd10 or Stk24/25 in mice led to decreased levels of Aqp2 protein in tubular epithelial cells, associated with impaired intracellular vesicle trafficking and increased expression of certain proteins. Treatment with Erlotinib normalized the expression level and membrane abundance of Aqp2 protein, partially rescuing the water reabsorption defect.

Stk24 protects against obesity-associated metabolic disorders by disrupting the NLRP3 inflammasome

Qiang Qin et al.

Summary: In this study, it was found that the expression of Stk24 is significantly downregulated in ATMs in obese individuals or those with type 2 diabetes, as well as in mice fed a high-fat diet. Stk24 deficiency exacerbates HFD-induced metabolic disorders and insulin insensitivity. Stk24 plays a role in regulating NLRP3 inflammasome assembly in ATMs, and its expression correlates negatively with body mass index and related indicators in human subjects.
