Bile acids and the gut microbiota: metabolic interactions and impacts on disease

Stephanie L. Collins et al.

Summary: This Review discusses host-microbiota interactions and their impact on bile acid metabolism and composition. The gut microbiota plays a crucial role in bile acid metabolism, with diverse biological roles for bile acids being discovered. External factors such as antibiotics and diet also influence bile acid composition. Understanding the impact of bile acid signaling networks in different organs is increasingly important for the development of targeted therapeutics.

Genetic manipulation of gut microbes enables single-gene interrogation in a complex microbiome

Wen-Bing Jin et al.

Summary: Understanding the causal contribution of microbiota genes to host biology is challenging due to the lack of genetic targeting for nonmodel gut commensals. A newly developed pipeline enables the identification of gene transfer methods and the construction of genetic tools for nonmodel microbes, facilitating the study of microbiota-host interactions.

Human gut bacteria produce TH17-modulating bile acid metabolites

Donggi Paik et al.

Summary: This study identifies bile acids produced by gut bacteria that inhibit the differentiation of T(H)17 cells, a key immune cell type involved in inflammatory disorders such as inflammatory bowel disease. The levels of these bile acids are reduced in patients with inflammatory bowel disease and are inversely correlated with the expression of T(H)17-cell-associated genes.

Identification and Characterization of Major Bile Acid 7α-Dehydroxylating Bacteria in the Human Gut

Kyung Hyun Kim et al.

Summary: In this study, a key group of 7 alpha-dehydroxylating bacteria responsible for converting primary bile acids (BAs) to secondary BAs in the human gut was identified through sequence similarity network, genome neighborhood network, and gene abundance analyses using human gut metagenomes. The abundance of this bacterial group was highly correlated with the occurrence of diverse diseases associated with bile acid 7 alpha-dehydroxylation. The metabolic features of this key bacterial group were characterized using their metagenome-assembled genomes. This approach is useful for identifying and characterizing key gut bacteria highly associated with human health and diseases.

A Gut-Restricted Lithocholic Acid Analog as an Inhibitor of Gut Bacterial Bile Salt Hydrolases

Arijit A. Adhikari et al.

Summary: Bile acids are essential in host physiology, with gut bacterial bile salt hydrolase (BSH) enzymes playing a key role in converting primary into secondary bile acids. The compound AAA-10 is a potent pan-BSH inhibitor, effectively suppressing BSH activity and modulating bile acid pool composition in vivo.

A bacterial bile acid metabolite modulates Treg activity through the nuclear hormone receptor NR4A1

Wei Li et al.

Summary: Bile acids regulate immune balance by affecting T cell differentiation, with isoalloLCA promoting T-reg cell differentiation through chromatin structure changes; gut bacteria capable of producing isoalloLCA belong to common human gut bacterial phylum Bacteroidetes, and NR4A1 is essential for isoalloLCA's effects on T-reg cells; significantly reduced levels of isoalloLCA in patients with inflammatory bowel diseases suggest a critical role for isoalloLCA and its bacterial producers in maintaining immune homeostasis in humans.

Dominant Bacterial Phyla from the Human Gut Show Widespread Ability To Transform and Conjugate Bile Acids

L. N. Lucas et al.

Summary: Gut bacteria play a critical role in modifying primary bile acids synthesized by the host, leading to the production of secondary bile acids that can regulate metabolic processes and influence gut microbiota composition. Through screening 72 bacterial isolates, this study identified 43 isolates capable of modifying unconjugated bile acids, expanding the known diversity of bacteria involved in bile acid transformations. The discovery of novel microbial conjugated bile acids and previously undescribed species highlights the importance of understanding the interactions between the microbiome, bile acid pool composition, and human health.

Degradation of Bile Acids by Soil and Water Bacteria

Franziska Maria Feller et al.

Summary: Bile acids are surface-active steroid compounds produced by vertebrates, serving as emulsifiers, signaling compounds, and antimicrobial barriers in the duodenum. When excreted into soil and water, bile acids provide carbon- and energy-rich growth substrates for diverse bacteria, predominantly studied in individual strains of specific genera.
