Preparation and Characterization of a Novel Multiparticulate Dosage Form Carrying Budesonide-Loaded Chitosan Nanoparticles to Enhance the Efficiency of Pellets in the Colon

Summary: In this study, chitosan nanoparticles loaded with budesonide were prepared using the ionic gelation technique to improve the delivery of orally administered nanoparticles to the colon. The size, zeta potential, encapsulation efficiency, shape, and drug release of the nanoparticles were investigated. The nanoparticles were then pelletized using the extrusion-spheronization method and coated with enteric and time-dependent polymers for colon-specific delivery. The formulations were tested for their anti-inflammatory effect in rats with induced colitis, and the relapse of colitis after treatment discontinuation was also monitored. The results showed that the pelletized budesonide nanoparticles released the drug steadily throughout the gastrointestinal tract, leading to long-lasting anti-inflammatory effects.

Summary: Kaolinite mineral was exfoliated into single nano-sheets (EXFK) and functionalized by methanol forming novel methoxy kaolinite nanosheet (MX/EXFK) as an efficient carrier for oxaliplatin drug (OXP). The functionalization of EXFK significantly improved the loading properties, achieving 433 mg/g (MX/EXFK) compared to 304.9 mg/g (EXFK). The release behavior of OXP from MX/EXFK exhibited continuous and controlled profiles, lasting up to 100 h (pH) and 180 h (phosphate buffer).

Its non-invasive delivery, ease of application, and safety profile contribute to its growing popularity. This review discusses the pharmacokinetics, pharmacodynamics, and safety profile of lipophilic drugs loaded in the NEG system, as well as its potential and future scope as a reliable delivery system.
