Prevalence of Plasmodium falciparum haplotypes associated with resistance to sulfadoxine-pyrimethamine and amodiaquine before and after upscaling of seasonal malaria chemoprevention in seven African countries: a genomic surveillance study

Khalid B. Beshir et al.

Summary: This study investigated the effectiveness of seasonal malaria chemoprevention in reducing parasite carriage among children in 13 countries in the Sahel region of Africa. The results showed a significant reduction in parasite carriage among children two years after the intervention scale-up. The findings suggest that seasonal malaria chemoprevention with sulfadoxine-pyrimethamine and amodiaquine remains effective, but the potential threat of resistance to sulfadoxine-pyrimethamine needs further monitoring.

Factors Influencing Second and Third Dose Observance during Seasonal Malaria Chemoprevention (SMC): A Quantitative Study in Burkina Faso, Mali and Niger

Anyirekun Fabrice Some et al.

Summary: This study aims to evaluate the factors influencing the adherence to the 2nd and 3rd doses of Amodiaquine (AQ) during seasonal malaria chemoprevention (SMC) in Burkina Faso, Mali, and Niger. The study found that the main causes of non-adherence were other illnesses and vomiting, while non-adherence related to parents was primarily due to their absence and traveling, and non-adherence related to community distributors was mainly due to missing the doses.

Identifying an optimal dihydroartemisinin-piperaquine dosing regimen for malaria prevention in young Ugandan children

Erika Wallender et al.

Summary: Intermittent preventive treatment with dihydroartemisinin-piperaquine (DP) is highly protective against malaria in children, with DP every 4 weeks associated with 95% protective efficacy. A PPQ level of 15.4 ng/mL reduces the malaria hazard by 95%, while malnutrition decreases PPQ exposure.

Balanced impacts of fitness and drug pressure on the evolution of PfMDR1 polymorphisms in Plasmodium falciparum

Marvin Duvalsaint et al.

Summary: The study found that WT N86/184F parasites were less fit than parasites with other haplotypes. Culturing with chloroquine favored 86Y mutant parasites, while culturing with lumefantrine favored N86 WT parasites. Parasites with double WT were in equilibrium with double mutant, but 86Y/Y184 overgrew N86/184F.

Effectiveness of seasonal malaria chemoprevention (SMC) treatments when SMC is implemented at scale: Case-control studies in 5 countries

Matthew Cairns et al.

Summary: Seasonal malaria chemoprevention (SMC) demonstrated high effectiveness against clinical malaria when implemented at scale through national malaria control programs, providing a very high level of personal protection, similar to the efficacy observed in clinical trials. The case-control design used in this study can be employed periodically by national malaria control programs to ensure the ongoing effectiveness of SMC treatments.
