SURF4-induced tubular ERGIC selectively expedites ER-to-Golgi transport

Rui Yan et al.

Summary: A highly elongated tubular ER-Golgi intermediate compartment (t-ERGIC) that selectively expedites the ER-to-Golgi transport for soluble cargoes of the receptor SURF4 has been discovered. The t-ERGIC has a distinctive elongated and thinned shape, and is marked by the small GTPases Rabl A/B instead of the canonical ERGIC marker ERGIC-53. It accelerates the trafficking of SURF4-bound cargoes with its large surface-to-volume ratio, high intracellular traveling speeds, and ER-Golgi recycling capabilities.

Summary: This study utilized whole-cell electron tomography to investigate the morphology, formation, and development of vacuoles in Arabidopsis stomatal lineage cells. The results showed that large vacuoles were generated from small vacuole fusion/maturation, and multivesicular body fusion played a role in stomatal vacuole formation.

Arabidopsis synaptotagmin 1 mediates lipid transport in a lipid composition-dependent manner

Tiantian Qian et al.

Summary: Endoplasmic reticulum-plasma membrane contact sites (EPCSs) in plant cells play a critical role in abiotic stress tolerance. Arabidopsis SYT1, enriched in EPCSs, has been shown to mediate lipid transfer between membranes. The lipid transfer activity of SYT1 is regulated by phosphatidylinositol 4,5-bisphosphate [PI(4,5)P-2] and Ca2+.

The ER Is a Common Mediator for the Behavior and Interactions of Other Organelles

Jaideep Mathur et al.

Summary: The optimal functioning of plant cells depends on efficient exchange between organelles, which may be facilitated by increased proximity and involvement of the endoplasmic reticulum (ER). This study found that under normal conditions, direct physical contact between organelles is not observed, but all organelles maintain close connectivity with the ER. Increased proximity between organelles occurs in response to stress, but the ER remains sandwiched between them. This provides evidence for the ER acting as a mediator in interactions between other organelles.

DeepContact: High-throughput quantification of membrane contact sites based on electron microscopy imaging

Liqing Liu et al.

Summary: In this study, a tool called DeepContact was developed for optimizing organelle segmentation and contact analysis using label-free electron microscopy (EM) through deep learning algorithms. DeepContact showed high efficiency and flexibility, capable of accommodating various organelle morphologies and identifying contacts of different widths. The study revealed previously unidentified coordinated rearrangements of organelles and a subtle wave of interaction between the endoplasmic reticulum (ER) and mitochondria in Sertoli cells during the seminiferous epithelial cycle, demonstrating the potential of DeepContact in bridging MCS dynamics to physiological and pathological processes.

Chaofan Chen et al.

Summary: Cell-to-cell communication in plants occurs through nanotubular passages called plasmodesmata, composed of endoplasmic reticulum and plasma membrane, associating with the actin-cytoskeleton. The coordination of these structures plays a crucial role in maintaining the architecture and conductivity of plasmodesmata, facilitating cell-to-cell communication in plant development and response to environmental stress.

Summary: Cellular versatility relies on accurate trafficking of proteins to their organelles, particularly through the secretory pathway which involves an elaborate network of interconnected tubules originating from the endoplasmic reticulum. Synaptotagmins at the endoplasmic reticulum-plasma membrane contact sites maintain diacylglycerol homeostasis during abiotic stress

Noemi Ruiz-Lopez et al.

Summary: Endoplasmic reticulum-plasma membrane contact sites (ER-PM CS) are essential for maintaining plasma membrane integrity under abiotic stresses in plant cells. The proteins SYT1 and SYT3 function as ER-PM tethers to preserve plasma membrane integrity. Cold stress can lead to increased accumulation of diacylglycerol at the plasma membrane, and SYT1 preferentially binds diacylglycerol in vivo.
