Glioma targeted therapy: insight into future of molecular approaches

Keyang Yang et al.

Summary: Gliomas are common brain tumors originating from glial cells. Treatment and prognosis differ significantly depending on the type of glioma. Traditional targeted therapy approaches have faced challenges, while new methods such as immunotherapy and the tumor microenvironment are gaining interest. More clinical trials are needed to evaluate the feasibility of personalized targeted therapy.

Calcium bursts allow rapid reorganization of EFhD2/Swip-1 cross-linked actin networks in epithelial wound closure

Franziska Lehne et al.

Summary: Calcium-dependent actin cross-linker EFhD2/Swip-1 is identified as a promoter of rapid actin network reorganization in epithelial wound closure. EFhD2/Swip-1 is highly upregulated in macrophages during metamorphosis and its loss or gain of function affects lamellipodial cell migration. TIRF analyses demonstrate that EFhD2/Swip-1 efficiently cross-links actin filaments in a calcium-dependent manner.

Precision Oncology in Lower-Grade Gliomas: Promises and Pitfalls of Therapeutic Strategies Targeting IDH-Mutations

Pasquale Persico et al.

Summary: This review examines the crucial role of isocitrate dehydrogenase (IDH) mutations in lower-grade gliomas and discusses different therapeutic strategies targeting these mutations. The study explores the physiological and pathological mechanisms by which IDH mutations contribute to tumorigenesis and presents two strategies: reducing intratumoral 2-HG levels and exploiting selective vulnerabilities caused by 2-HG accumulation. The review critically evaluates preclinical evidence and available clinical data of mutant-selective IDH inhibitors and IDH-targeted vaccines, as well as the rationale of utilizing 2-HG-related epigenetic and metabolic weaknesses as an alternative approach.

Time to focus on circulating nucleic acids for diagnosis and monitoring of gliomas: A systematic review of their role as biomarkers

Ana Belen Diaz Mendez et al.

Summary: Gliomas are tumors that originate from progenitors within the central nervous system, with a majority requiring surgical intervention and having a tendency to recur. Imaging has limitations in monitoring tumor burden post-surgery, and liquid biopsies show promise in aiding diagnosis and prognosis.

Key proteins of invadopodia are overexpressed in oral squamous cell carcinoma suggesting an important role of MT1-MMP in the tumoral progression

Geovanni Pereira Mitre et al.

Summary: The immunoexpression of invadopodia-related proteins (TKs5, cortactin, TKs4 and MT1-MMP) is significantly increased in oral squamous cell carcinoma (OSCC), with overexpression of MT1-MMP potentially serving as a promising prognostic marker for OSCC.

Spatial Dissection of Invasive Front from Tumor Mass Enables Discovery of Novel microRNA Drivers of Glioblastoma Invasion

Yulun Huang et al.

Summary: The study demonstrates a strategy to isolate invasive and tumor core cells from highly invasive patient-derived models, revealing differences in invasion capacity and miRNA expression. Silencing top miRNA candidates successfully blocks invasion, while targeting a common gene with inhibitors suppresses invasion. In vivo treatment effectively eliminates GBM invasion and extends animal survival, highlighting the importance of accurate starting materials in understanding cancer invasion.

Noncoding RNAs in Glioblastoma: Emerging Biological Concepts and Potential Therapeutic Implications

Uswa Shahzad et al.

Summary: Noncoding RNAs (ncRNAs) have emerged as a novel class of genomic regulators, ushering in a new era in molecular biology. With the advent of advanced genetic sequencing technology, several different classes of ncRNAs have been uncovered, including microRNAs (miRNAs), long noncoding RNAs (lncRNAs), circular RNAs (circRNAs), and piwi-interacting RNAs (piRNAs), which have been linked to many important developmental and disease processes and are being pursued as clinical and therapeutic targets.

EFhd2 brain interactome reveals its association with different cellular and molecular processes

Ahlam S. Soliman et al.

Summary: EFhd2 is a highly expressed calcium-binding protein in the central nervous system that interacts with tau protein, a key pathological hallmark in Alzheimer's disease and related dementias. This study reveals an EFhd2 brain interactome associated with various cellular and molecular processes, suggesting its involvement in modulating these processes in both physiological and pathological conditions of the nervous system. The identification of proteins associated with cytoskeleton components, vesicle trafficking, calcium signaling, stress response, and metabolic pathways provides insights into the potential roles of EFhd2 in the brain.
