Piperlongumine synergistically enhances the antitumour activity of sorafenib by mediating ROS-AMPK activation and targeting CPSF7 in liver cancer

Liyun Zheng et al.

Summary: Sorafenib is a first-line agent for advanced liver cancer, but drug resistance affects its efficacy. In this study, we demonstrated that piperlongumine (PL) enhances the cytotoxicity of sorafenib in liver cancer cells by inducing ROS generation, mitochondrial dysfunction, and apoptosis. Activation of the AMPK signaling pathway and disruption of RNA processing are involved in the antitumour activity of PL and sorafenib combination treatment.

Oxidative Damage to Mitochondria Enhanced by Ionising Radiation and Gold Nanoparticles in Cancer Cells

Farnaz Tabatabaie et al.

Summary: Gold nanoparticles (AuNP) can enhance the efficacy of radiation therapy by increasing cellular stress and cell killing. This study demonstrates that the inclusion of AuNPs in radiation therapy leads to increased mitochondrial stress and reactive oxygen species (ROS) production, potentially resulting in more effective killing of cancer cells.

Application of glutathione depletion in cancer therapy: Enhanced ROS-based therapy, ferroptosis, and chemotherapy

Boyi Niu et al.

Summary: Glutathione (GSH) plays a crucial role in cellular antioxidative system, particularly in cancer cells, where a high level of GSH is essential for scavenging ROS and detoxifying xenobiotics. Depletion of GSH has been shown to enhance the efficacy of various therapies such as ROS-based therapy, ferroptosis, and chemotherapy. This review comprehensively discusses the importance of GSH depletion in cancer therapy and various strategies employed to achieve it.

Single-cell profiling of myeloid cells in glioblastoma across species and disease stage reveals macrophage competition and specialization

Ana Rita Pombo Antunes et al.

Summary: By utilizing single-cell RNA sequencing and CITE-seq, this study reveals the complexity of the immune landscape in glioblastomas, including conserved populations of dendritic cells and macrophages. The results indicate that microglia-derived tumor-associated macrophages dominate in newly diagnosed tumors, but are replaced by monocyte-derived TAMs in recurrences.

Summary: Researchers successfully established a highly reproducible mouse GBM surgical model that faithfully recapitulates the rapid recurrence of human GBM after surgery. By implanting G422(TN)-GBM cells, they found that surgery was an effective therapy but not a curable one.

Immunostimulatory bacterial antigen-armed oncolytic measles virotherapy significantly increases the potency of anti-PD1 checkpoint therapy

Summary: This study found that combination NAP-immunovirotherapy induced massive influx of lymphoid cells in mouse brain, with CD8+ T cell predominance; therapeutic efficacy was CD8+ T cell dependent. Inhibition of the IFN response pathway using the JAK1/JAK2 inhibitor ruxolitinib decreased PD-L1 expression on myeloid-derived suppressor cells in the brain and further potentiated the therapeutic effect of MV-s-NAP-uPA and anti-PD1.
