Circular RNAs in cholangiocarcinoma

Wenwei Liao et al.

Summary: Cholangiocarcinoma (CCA) is a primary biliary malignancy with a poor prognosis and limited treatment options. Recently, circular RNAs (circRNAs) have gained attention as potential therapeutic targets and prognostic biomarkers for CCA. Dysregulated circRNAs play vital roles in the development of CCA through various mechanisms, including disruption of miRNA regulation, interaction with RNA-binding proteins, activation of signaling pathways, and modulation of proteins and peptides. Inflammatory microenvironment and chemotherapy resistance are also influenced by circRNAs in CCA. Therefore, circRNAs are crucial for early diagnosis and prediction of CCA, and further research is needed to understand their roles and mechanisms in CCA.

The emerging roles of circRNAs in cancer and oncology

Lasse S. Kristensen et al.

Summary: Circular RNAs (circRNAs) are a novel class of primarily non-coding RNAs with increasingly recognized roles in cancer development and progression through diverse mechanisms of action. In this review, the authors summarize the current understanding of circRNA biogenesis, regulation, physiological functions and pathophysiological roles in cancer. They also discuss the clinical potential of circRNAs as biomarkers, therapeutic agents and drug targets in oncology, as well as the research controversies, technical issues, and biological knowledge gaps that need to be addressed.

Circular RNAs: Characterization, cellular roles, and applications

Chu-Xiao Liu et al.

Summary: In this review, we discuss the methods used in circular RNA studies and the current understanding of their regulatory roles and potential applications. Circular RNAs can modulate transcription and splicing, regulate the stability and translation of mRNAs, interfere with signaling pathways, and play important roles in cellular processes and immune responses.

Third-generation EGFR and ALK inhibitors: mechanisms of resistance and management

Alissa J. Cooper et al.

Summary: Patients with non-small-cell lung cancers (NSCLCs) harboring oncogenic EGFR or ALK alterations can benefit from therapies targeting these alterations, although acquired resistance to these agents is common. Third-generation inhibitors have extended the response durations of many patients with NSCLCs harboring these alterations, albeit with differing patterns of resistance to those associated with earlier-generation agents.

Companion Diagnostics and Predictive Biomarkers for MET-Targeted Therapy in NSCLC

Jan Trost Jorgensen et al.

Summary: MET is a receptor tyrosine kinase that plays a significant role in cancer cell progression and is targeted by several drugs under development for the treatment of different cancers. However, the lack of effective biomarkers to select the right patients for treatment may be one of the reasons why relatively few of these drugs have shown sufficient clinical activity and obtained regulatory approval. MET exon 14 skipping mutation and MET amplification detected by FISH are two potential biomarkers, with the former showing sufficient predictive properties and the latter showing promising results in the treatment of NSCLC patients.

Summary: The study identified two factors, SFPQ and NONO, enriched around circRNA loci, particularly playing important roles in DALI circRNAs. Results indicate that SFPQ plays a crucial role in maintaining intron integrity, while also revealing novel features governing Alu-independent circRNA production.

Comprehensive profiling of circular RNAs with nanopore sequencing and CIRI-long

Jinyang Zhang et al.

Summary: This study presents a method for enrichment and full-length sequencing of circRNA isoforms using nanopore technology, allowing high-throughput detection and reconstruction of circRNAs. The developed algorithm, CIRI-long, accurately reconstructs the sequence of circRNAs from long-read sequencing data. Analysis of adult mouse brain samples identified diverse circRNA types, including a novel intronic self-ligated circRNA with special splicing and expression patterns.

A comprehensive characterization of the cell-free transcriptome reveals tissue- and subtype-specific biomarkers for cancer detection

Matthew H. Larson et al.

Summary: Cell-free RNA is a promising analyte for cancer detection, with the potential to identify tissue- and subtype-specific biomarkers in breast and lung cancer patients. The presence of dark channel biomarker (DCB) genes in cfRNA from individuals with cancer suggests they could enhance cancer detection in patients with low levels of circulating tumor DNA.

Nanopore sequencing of brain-derived full-length circRNAs reveals circRNA-specific exon usage, intron retention and microexons

Karim Rahimi et al.

Summary: Circular RNAs, a class of covalently joined non-coding RNAs, play functional roles in cellular processes and exhibit overlap with exons in linear mRNAs. Long-read sequencing techniques have been used to characterize circRNAs, revealing a large number of splicing events and proposing altered exon usage as a resistance mechanism to nonsense-mediated decay. The development of targeted long-read sequencing methods offers a comprehensive approach to understanding the structure and function of circRNAs.
