However, a comprehensive review on the structure-activity relationships and dose-dependent effects of HMOs is lacking. This review provides an overview of the structures and synthetic pathways of HMOs, and summarizes the identified structure-function relationships. It also highlights recent advances in understanding the interactions between health benefits and dosage effects based on cellular tests and animal experiments.

Transglycosylation Activity of Engineered Bifidobacterium Lacto-N-Biosidase Mutants at Donor Subsites for Lacto-N-Tetraose Synthesis

Summary: This study focused on mutational strategies in the donor subsites of lacto-N-biosidase to reduce hydrolytic activity and enable transglycosylation for the synthesis of LNT. Mutants with residual hydrolase activity between 0.05% to 1.6% of the wild-type enzyme showed transglycosylating enzymes with LNT yields ranging from 10-30%. Mutations in Trp394 had a significant impact on the transglycosylation/hydrolysis ratio, with the W394F mutant demonstrating the highest biocatalytic efficiency in producing LNT at a yield of 32%.
