Biallelic GINS2 variant p.(Arg114Leu) causes Meier-Gorlin syndrome with craniosynostosis

Maria J. Nabais Sa et al.

Summary: The study identified a novel disease-associated gene GINS2 in Meier-Gorlin syndrome (MGORS), expanding the genetic etiology of the disorder. A novel homozygous missense variant in GINS2 was identified through exome sequencing, leading to increased sensitivity to nicotinamide and potential disruption of the effective interaction between the GINS complex and CDC45.

Summary: The International Union of Immunological Societies Expert Committee has published an updated classification of inborn errors of immunity, which includes 55 novel monogenic gene defects and 1 phenocopy caused by autoantibodies. This update contributes to our understanding of the molecular, cellular, and immunological mechanisms of human immune diseases, and provides a resource for molecular diagnosis of heritable immunological disorders and scientific research on monogenic and related human immune diseases.

Partial loss-of-function mutations in GINS4 lead to NK cell deficiency with neutropenia

Matilde I. Conte et al.

Summary: Human NK cell deficiency (NKD) is a primary immunodeficiency disease characterized by missing or dysfunctional NK cells. This study describes a familial NKD case with compound heterozygous variants in the GINS4 gene, which is involved in NK cell development. The study demonstrates that these mutations have a damaging impact on the expression and assembly of the GINS complex, leading to delayed cell cycle progression in NK cells. These findings highlight the importance of the GINS4 gene in NK cell biology and provide insights into the underlying mechanisms of NKD.

Single-cell profiling reveals the trajectories of natural killer cell differentiation in bone marrow and a stress signature induced by acute myeloid leukemia

Adeline Crinier et al.

Summary: NK cells in human bone marrow can be classified into three subpopulations, with NK0 cells believed to be precursors of circulating CD56(dim) NK1-like NK cells and CD56(bright) NK2-like NK cells under physiological conditions. Transcriptomic analysis showed stress-induced repression of NK cell effector functions in bone marrow NK cells from patients with acute myeloid leukemia (AML), with reduced levels of CD160, but higher survival rates in the CD160(high) group.

MCM complex members MCM3 and MCM7 are associated with a phenotypic spectrum from Meier-Gorlin syndrome to lipodystrophy and adrenal insufficiency

Karen M. Knapp et al.

Summary: The MCM2-7 helicase plays essential roles in DNA replication and its disruption is associated with rare disorders like Meier-Gorlin syndrome and immune deficiency. This study identified pathogenic variants in MCM7 and MCM3, confirming their impact on DNA replication. The diverse phenotypes observed in patients with similar variant profiles in MCM7 highlight the complexity of disrupted MCM function.

Bi-allelic MCM10 variants associated with immune dysfunction and cardiomyopathy cause telomere shortening

Ryan M. Baxley et al.

Summary: MCM10 deficiency results in chronic replication stress, reducing cell viability due to increased genomic instability and telomere erosion. Loss of MCM10 function constrains telomerase activity, potentially leading to a build-up of abnormal replication structures and terminally-arrested replication forks that require processing by MUS81.
