Detection of driver mutations in plasma cell-free nucleic acids in differentiated thyroid neoplasm

Susmita Dutta et al.

Summary: This proof-of-concept study shows that driver mutations in differentiated thyroid tumors can be detected in plasma, with a detection rate of up to 80% in malignant nodules. The use of an 8-gene panel can also predict cancer subtypes. There is strong agreement between driver mutations found in plasma samples, FNAC materials, and histopathology samples, indicating the potential for this noninvasive diagnostic tool in preoperative assessment and tumor recurrence detection.

Characteristics of PTEN Mutation in Thyroid Tumours: A Retrospective Chart Review

Saruchi Bandargal et al.

Summary: This study investigated the relationship between PTEN mutations and thyroid malignancy, and whether these malignancies are aggressive. The results showed that 37.5% of PTEN-mutated thyroid nodules were malignant, and aggressive features were present in 33.33% of the malignant tumors. It is hypothesized that PTEN-mutated thyroid nodules can acquire high allele frequency and widespread copy number alterations over time, which may worsen the effects of PTEN mutations.

The 5th edition of WHO classification of tumors of endocrine organs: changes in the diagnosis of follicular-derived thyroid carcinoma

Fulvio Basolo et al.

Summary: The 5th edition of the WHO classification of endocrine tumors, released in 2022, introduces several novelties in the nomenclature and histopathological diagnosis of follicular-derived thyroid neoplasms. Tumor types are now categorized into benign tumors, low-risk neoplasms, and malignant neoplasms based on prognostic risk. A grading system for differentiated thyroid carcinomas has been implemented for better tumor stratification. The review critically discusses the practical effects on diagnostic pathology and the expected clinical implications of the new classification scheme.
