Exploring the Use of Gas Chromatography Coupled to Chemical Ionization Mass Spectrometry (GC-CI-MS) for Stable Isotope Labeling in Metabolomics

Jordi Capellades et al.

Summary: Isotopic-labeling experiments are crucial for monitoring metabolic flux in biological systems, with GC methods coupled to CI-MS showing clear advantages in providing precise analysis of metabolic intermediates. The experimental determination of metabolic fluxes can be inferred from the rearrangement of stable isotope tracers and can be detected by mass spectrometry.

The Source of Glycolytic Intermediates in Mammalian Tissues

Tara TeSlaa et al.

Summary: Glycolysis is central to organismal metabolism, with circulating glucose and intracellular glycogen being the main sources of glycolytic intermediates. Gluconeogenesis also contributes to glycolysis to a lesser extent. Different tissues show variations in utilizing circulating glucose for glycolysis, with most tissues relying on glycogen breakdown for glycolytic intermediates. Overall, the body slowly breaks down internal glycogen stores while select tissues rapidly catabolize circulating glucose for oxidation.

Targeted Profiling of Short-, Medium-, and Long-Chain Fatty Acyl-Coenzyme As in Biological Samples by Phosphate Methylation Coupled to Liquid Chromatography-Tandem Mass Spectrometry

Peng Li et al.

Summary: Fatty acyl-coenzyme As (acyl-CoAs) are essential in lipid metabolism pathways, with short-chain acyl-CoAs being part of metabolomics and medium- to (very) long-chain acyl-CoAs being the focus of lipidomics studies. A derivative strategy based on phosphate methylation was proposed in this work, enabling full coverage and good peak shape in liquid chromatography. This method was successfully validated in cultured cells and applied to analyze distinct acyl-CoA profiles in HeLa cells and platelets of coronary artery disease patients.
