ID-MAM: A Validated Identity and Multi-Attribute Monitoring Method for Commercial Release and Stability Testing of a Bispecific Antibody

Adam R. Evans et al.

Summary: Post-translational modifications (PTMs) can impact the safety and efficacy of protein therapeutics, making them critical quality attributes (CQAs) that need to be controlled for product quality. Peptide mapping with online mass spectrometry has been used for years to monitor PTM CQAs during product development, but implementing high-resolution methods in GMP-compliant commercial QC labs can be challenging. A targeted multi-attribute monitoring (MAM) method using low-resolution mass spectrometry has been developed and validated for identity testing, sequence variant control, and CQA quantitation in commercial QC labs. This method has been successfully validated in multiple labs across countries for commercial release and stability testing of a bispecific antibody.
