Dynamic Heterogeneity at the Interface of an Intrinsically Disordered Peptide

Summary: This study investigates the dynamic properties of water near the α-synuclein protein associated with Parkinson's disease. The results show that the translational and rotational mobility of water molecules near the peptide segments are significantly restricted, with water near the hydrophobic segment exhibiting more restricted diffusivity. The time scales of peptide-water and water-water hydrogen bond relaxations correlate with the diffusion of interfacial water molecules. The hindered dynamic environment near a specific segment can enhance early stages of peptide aggregation.

Adsorption Energetics of Amino Acid Analogs on Polymer/Water Interfaces Studied by All-Atom Molecular Dynamics Simulation and a Theory of Solutions

Summary: The energetics of adsorption on the interfaces of PMEA, PMMA, and PBA with water were studied using all-atom molecular dynamics simulation. The adsorption free energy was found to be favorable for all combinations of solute and polymer, and PMMA exhibited the strongest adsorption for hydrophobic solutes while PMEA showed the strongest adsorption for hydrophilic solutes. The polymer contribution to the adsorption free energy was larger with increasing solute size. The total free energy of adsorption correlated well with the solvation free energy in bulk water for hydrophobic solutes only.

Molecular Structure and Vibrational Spectra of Water Molecules Sorbed in Poly(2-methoxyethylacrylate) Revealed by Molecular Dynamics Simulation

Summary: Molecular dynamics simulations reveal that water molecules tend to form hydrogen bonds with the carbonyl oxygen and methoxy oxygen of the PMEA side chains, enhancing the head-to-tail stacking structure. Further penetration of water into the PMEA structure gradually increases water-water hydrogen bonding, promoting the formation of water clusters even below the equilibrium water content.

Effects of Side-Chain Spacing and Length on Hydration States of Poly(2-methoxyethyl acrylate) Analogues: A Molecular Dynamics Study

Summary: The hydration states of polymers have a direct impact on biomolecule adsorption. Experimental studies on poly(2-methoxyethyl acrylate) analogues with different side-chain spacings and lengths showed that HPx has more IW content but lower nonthrombogenicity compared to PMCyA. Molecular dynamics simulations revealed that more methylene groups in the side chain hinder water penetration, resulting in lower IW content in PMCyA.

Summary: Improving hydrophilicity is crucial for enhancing the biocompatibility of polymer surfaces. Surprisingly, the more hydrophobic PMEA surfaces exhibit better biocompatibility compared to the more hydrophilic PHEMA surfaces. Molecular dynamics simulations and analysis of RDF and B-2 values confirm that the lower affinity of MEA trimers with nonpolar foulants contributes to the excellent biocompatibility of PMEA.
