Protein Expression of the Microglial Marker Tmem119 Decreases in Association With Morphological Changes and Location in a Mouse Model of Traumatic Brain Injury

Domenico Mercurio et al.

Summary: The study suggests that after acute brain injuries, a decrease in Tmem119 expression in reactive microglia may be due to the process of microglial activation, which involves a shrinking of their branchings to acquire an ameboid shape. The Tmem119 immunoreactivity decreases in reactive microglia to similar levels as blood-borne macrophages, thus failing to differentiate the two myeloid populations.

Fenghui Ye et al.

Summary: Using a Tmem119-EGFP transgenic mouse model, the study found that the number of perihematomal microglia significantly decreased on day 1 after ICH, but markedly increased by day 3 and day 7. Injection of iron or thrombin induced microglia death or proliferation, respectively.

The Role of Thrombin in Brain Injury After Hemorrhagic and Ischemic Stroke

Fenghui Ye et al.

Summary: Thrombin levels in the brain increase after stroke, contributing to brain pathology through harmful and protective mechanisms. Thrombin can affect brain injury via multiple pathways, linking the coagulation system to the immune system and potentially leading to short-term and long-term functional deficits.

Role of lipocalin-2 in extracellular peroxiredoxin 2-induced brain swelling, inflammation and neuronal death

Summary: This study found that intracerebral injection of PRX-2 upregulates LCN-2 protein expression, and LCN-2 plays a crucial role in PRX-2 induced neuroinflammation and brain damage. Additionally, the presence of LCN-2 attenuates the neurological deficits caused by PRX-2.

Temporally distinct myeloid cell responses mediate damage and repair after cerebrovascular injury

Panagiotis Mastorakos et al.

Summary: Positive vascular recanalization after endovascular thrombectomy may lead to cerebral edema and worsened clinical outcomes, especially in patients with hemorrhagic transformation. Resident microglia initially stabilize damaged vessels, followed by an influx of myelomonocytic cells causing severe edema. Prolonged blockade of myeloid cell recruitment can prevent severe edema but also interfere with vascular repair.

Microglia-derived interleukin-10 accelerates post-intracerebral hemorrhage hematoma clearance by regulating CD36

Qian Li et al.

Summary: IL-10 regulates microglial phagocytosis and monocyte-derived macrophage infiltration after ICH, with CD36 identified as a key phagocytosis effector regulated by IL-10. Leveraging IL-10 signaling may provide a useful strategy for accelerating hematoma clearance and improving neurologic outcome.

Brain injury instructs bone marrow cellular lineage destination to reduce neuroinflammation

Samuel X. Shi et al.

Summary: Acute brain injury can mobilize circulating leukocytes to aggravate neural injury. This study demonstrates that bone marrow hematopoietic stem cells (HSCs) may be skewed towards the myeloid lineage in patients with intracerebral hemorrhage, leading to increased production of Ly6C(low) monocytes that suppress neuroinflammation and brain injury. The brain injury-induced modulation of HSC lineage development highlights the potential therapeutic effects of targeting bone marrow in neuroimmune interactions to reduce brain inflammation.

Divergent Functions of Tissue-Resident and Blood-Derived Macrophages in the Hemorrhagic Brain

Che-Feng Chang et al.

Summary: Our study shows that microglia and MDMs have distinct functional properties in the brain after ICH, with MDMs exhibiting higher phagocytic activity, erythrophagocytosis, and antigen-presenting capabilities compared to microglia. This suggests that the different origins of microglia and MDMs lead to divergent responses and functions in the inflamed brain following ICH.

CD47 blocking antibody accelerates hematoma clearance and alleviates hydrocephalus after experimental intraventricular hemorrhage

Fenghui Ye et al.

Summary: Blocking CD47 in the hematoma accelerated hematoma clearance and alleviated hemolysis and hydrocephalus development after intraventricular hemorrhage, suggesting CD47 might be valuable in the future treatment for IVH. Erythrophagocytosis was observed as soon as 4 h after IVH and was enhanced on day 3. CD47 blockade also upregulated intra-hematoma infiltration of CD68, heme oxygenase-1 and ferritin positive phagocytes by day 3.
