Hepatic stellate cells in the injured liver: Perspectives beyond hepatic fibrosis

Devaraj Ezhilarasan

Summary: The understanding of the pathological role of hepatic stellate cells (HSCs) in liver diseases has significantly increased over the past two decades, with a focus not only on liver fibrosis but also on conditions like cholestasis, portal hypertension, and liver cancer progression. Activated HSCs play a critical role in secreting extracellular matrix products and impacting the tumor microenvironment, leading to tumor cell proliferation, metastasis, and immune suppression. The activation of HSCs in the damaged liver is a crucial process involved in the development of cholestasis, portal hypertension, and liver cancer.

Review of galectin-3 inhibitors in the treatment of nonalcoholic steatohepatitis

Atef Al Attar et al.

Summary: Gal-3 is a beta-galactoside binding protein associated with disease pathologies such as chronic inflammation and fibrogenesis, with ongoing studies investigating its role in NASH. Gal-3 inhibitors, particularly Belapectin, have shown promising results in NASH patients with advanced fibrosis, with further research aiming to replicate positive findings and demonstrate greater efficacy. If proven effective, Belapectin may be used in combination with other drugs targeting steatohepatitis for maximum efficacy and disease reversal.

Cell and Tissue Therapy for the Treatment of Chronic Liver Disease

Yaron Bram et al.

Summary: Liver disease is a significant clinical issue affecting 600 million people worldwide. Liver transplantation, although the gold standard treatment for liver failure, currently meets less than 10% of the global demand due to a shortage of available organs. Cell- and tissue-based therapies offer a promising alternative to organ transplantation.

Summary: NAFLD and NASH are leading chronic liver diseases worldwide, causing progressive liver injury that can lead to cirrhosis and hepatocellular carcinoma. These diseases are associated with hepatic glucose and lipid metabolism, and are influenced by genetic, epigenetic, and environmental factors.

Summary: High expression of Galectin-1 and -3 is associated with worse outcomes in hepatocellular carcinoma patients, while high expression of Galectin-4 and -9 is associated with better outcomes. Serum Galectin-3 and -9 levels are positively associated with the risk of chronic liver diseases.

Galectin-3 facilitates the proliferation and migration of nasopharyngeal carcinoma cells via activation of the ERK1/2 and Akt signaling pathways, and is positively correlated with the inflammatory state of nasopharyngeal carcinoma

Mei Li et al.

Summary: In patients with nasopharyngeal carcinoma, Gal-3 expression is upregulated and positively correlated with the inflammatory state of the cancer. Gal-3 promotes proliferation and migration of NPC cells while inhibiting apoptosis. The activation of ERK1/2 and Akt may be the underlying mechanism for the effects of Gal-3 on NPC.

Galectin-3 N-terminal tail prolines modulate cell activity and glycan-mediated oligomerization/ phase separation

Zihan Zhao et al.

Summary: Gal-3's N-terminal tail with proline-rich residues plays a crucial role in mediating various cellular activities, affecting cell migration, activation, endocytosis, and hemagglutination. The research also reveals the significant impact of prolines in regulating Gal-3 oligomerization process.
