Summary: Research findings indicate that SUMO protease SENP1 promotes T cell memory development through Sirt3 deSUMOylation and enhances mitochondrial fusion and OXPHOS. Additionally, the glycolytic intermediate fructose-1,6-bisphosphate (FBP) acts as a negative regulator that suppresses AMPK-mediated activation of the SENP1-Sirt3 axis, thereby decreasing T cell memory development. Glucose limitation activates AMPK and subsequent SENP1-Sirt3 signaling to promote T cell memory development. Memory T cells heavily rely on fatty acid oxidation, which is controlled by AMPK sensing of glucose scarcity triggering SENP1-Sirt3 signaling for mitochondrial fusion and memory differentiation.
