Recent genetic exchanges and admixture shape the genome and population structure of the zoonotic pathogen Cryptosporidium parvum

Giulia Corsi et al.

Summary: This study compares whole genome sequences of Cryptosporidium parvum isolates from human and ruminant hosts collected from different regions globally. It finds evidence of genetic exchanges between different host species and geographic origins, with these exchanges potentially increasing the parasite's virulence. The study suggests that globalization and close human-animal contacts provide opportunities for genetic exchanges between previously isolated parasite lineages, which can impact host-parasite coevolution dynamics.

Structure, sequon recognition and mechanism of tryptophan C-mannosyltransferase

Joel S. Bloch et al.

Summary: C-linked glycosylation is essential for the trafficking, folding, and function of secretory and transmembrane proteins involved in cellular communication processes. Cryogenic-electron microscopy structures of Caenorhabditis elegans CMT reveal how the CMT recognizes the glycosylation sequon and triggers conformational activation of the donor substrate, enabling C-glycosylation of proteins. These findings have implications for understanding human diseases and potential therapeutic targeting of specific CMT paralogs.

Global Population Genomics of Two Subspecies of Cryptosporidium hominis during 500 Years of Evolution

Swapnil Tichkule et al.

Summary: This study presents a whole-genome study of Cryptosporidium hominis, a major cause of diarrhea in low- and middle-income countries. The study identifies two distinct lineages of C. hominis, with one lineage predominantly found in low-income countries in Africa and Asia, and another lineage found in high-income countries in Europe, North America, and Oceania. The study also detects genomic regions of introgression between the lineages, leading to high diversity and divergence in potential virulence genes.

Tryptophan C-mannosylation is critical for Plasmodium falciparum transmission

Sash Lopaticki et al.

Summary: This study demonstrates the importance of tryptophan C-mannosylation by the DPY19 enzyme in stabilizing TSR proteins and enabling successful transmission of the human malaria parasite to mosquitoes. Deficiency in DPY19 disrupts C-glycosylation, leading to defects in gametocyte egress, exflagellation, and oocyst formation, ultimately impairing mosquito infection.

Protein C-Mannosylation and C-Mannosyl Tryptophan in Chemical Biology and Medicine

Shiho Minakata et al.

Summary: C-mannosylation is a post-translational modification of proteins in the endoplasmic reticulum, playing important roles in protein folding, sorting, and secretion. The degradation product of C-mannosylated proteins, C-Man-Trp, is upregulated in blood of patients with renal dysfunction, potentially involved in human kidney diseases. Dysregulation of protein C-mannosylation may be associated with the pathophysiology of human diseases.

What Are We Missing by Using Hydrophilic Enrichment? Improving Bacterial Glycoproteome Coverage Using Total Proteome and FAIMS Analyses

Ameera Raudah Ahmad Izaham et al.

Summary: This study explores alternative approaches to studying bacterial glycoproteomes without relying on HILIC enrichment. By analyzing Burkholderia species, it was found that short aliphatic O-linked glycopeptides are absent in HILIC enrichments but can be identified in whole proteome samples. High-field asymmetric waveform ion mobility spectrometry (FAIMS) fractionation allowed for the enrichment of these glycopeptides from complex samples, expanding the observable glycoproteome of Burkholderia species by at least 25%. Additionally, the applicability of FAIMS for glycopeptide enrichment was demonstrated with N-linked glycopeptides from Campylobacter fetus subsp. fetus.
