Molecular origins of reduced activity and binding commitment of processive cellulases and associated carbohydrate-binding proteins to cellulose III

Summary: This study investigates the structural differences between cellulose I and cellulose III allomorphs and their impact on enzymatic hydrolysis efficiency. By monitoring enzyme-cellulose interactions using single-molecule force spectroscopy, it was found that reduced binding affinity of Cel7A on cellulose III leads to slower enzymatic activity. The findings suggest the use of simpler single-site models to explain the binding mechanisms between cellulolytic enzymes and cellulose.
