Uncovering Zn2+as a cofactor of FAD-dependent Pseudomonas aeruginosa PAO1 D-2-hydroxyglutarate dehydrogenase

Summary: Pseudomonas aeruginosa uses D-2-hydroxyglutarate dehydrogenase (PaD2HGDH) to couple the oxidation of D-2-hydroxyglutarate (D2HG) to L-serine biosynthesis for survival. Knockout of PaD2HGDH hinders the growth of P. aeruginosa, making it a potential therapeutic target. This study investigates the metal composition and role of metal as an activator or cofactor for PaD2HGDH.

Kinetic solvent viscosity effects uncover an internal isomerization of the enzyme-substrate complex in Pseudomonas aeruginosa PAO1 NADH: Quinone oxidoreductase

Summary: NAD(P)H: quinone oxidoreductases (NQOs) play a crucial role in detoxifying quinones as antioxidants in both Prokaryotes and Eukaryotes. This study investigates the kinetics of NQO from Pseudomonas aeruginosa PAO1 in the reduction of benzoquinone and juglone. The results suggest that flavin reduction is the rate-limiting step for overall turnover, and the hydride transfer from NADH to FMN and quinol product release partially limit the turnover. Furthermore, the study demonstrates how the properties of quinol substrates can affect the equilibrium of NQO esomerization.

Effects of free soluble iron on thermal aggregation of hemoglobin

Summary: Iron ions, both in Fe2+ and Fe3+ forms, were found to inhibit hemoglobin aggregation, with a 60% inhibition in the presence of 100 mg/L FeCl3. Other metal ions such as Cu2+, Zn2+, and Ni2+ did not have significant effects on hemoglobin aggregation. These findings suggest a chaperone-like behavior of free iron that could have important consequences on the severity of hematological diseases.
