Genetic analyses identify GSDMB associated with asthma severity, exacerbations, and antiviral pathways

Xingnan Li et al.

Summary: Multiple SNPs in the gasdermin B (GSDMB) gene are associated with asthma severity and longitudinal asthma exacerbations. Higher expression levels of GSDMB correlate with asthma exacerbations and pathways related to IFN signaling and immune system. GSDMB is identified as a functional gene for both asthma susceptibility and severity.

Prioritization of candidate causal genes for asthma in susceptibility loci derived from UK Biobank

Kim Valette et al.

Summary: The study identified 72 asthma-associated loci and potential therapeutic targets through GWAS, TWAS, and Mendelian randomization. Blood showed the largest fold enrichment of regulatory and functional annotations among asthma-associated variants. Among 485 blood eQTL-regulated genes associated with asthma, 50 were confirmed to be causal.

A cross-population atlas of genetic associations for 220 human phenotypes

Saori Sakaue et al.

Summary: The study conducted deep-phenotype genome-wide association studies in BioBank Japan, identifying approximately 5,000 new loci and improving the resolution of the genomic map of human traits. By decomposing matrices of phenome-wide summary statistics, responsible variants and biological mechanisms underlying current disease classifications across populations were pinpointed.

Airway mucin MUC5AC and MUC5B concentrations and the initiation and progression of chronic obstructive pulmonary disease: an analysis of the SPIROMICS cohort

Giorgia Radicioni et al.

Summary: This study investigated the relative contribution of MUC5AC and MUC5B to airways disease in COPD, revealing increased MUC5AC concentration as a potential biomarker for COPD initiation, progression, and exacerbation risk. MUC5AC concentration showed greater sensitivity to COPD severity and could be a target for future therapeutic strategies.

Genetic and non-genetic factors affecting the expression of COVID-19-relevant genes in the large airway epithelium

Silva Kasela et al.

Summary: This study reveals that variation in the expression of COVID-19-related genes is associated with host factors, environmental exposures, and likely host genetic variation. ACE2 gene expression is linked to risk factors like active smoking, obesity, and hypertension, while an association with interferon-related inflammation is driven by the truncated, non-binding ACE2 isoform.
