Insights into 4-hydroxyphenylpyruvate dioxygenase-inhibitor interactions from comparative structural biology

Hong-Yan Lin et al.

Summary: 4-Hydroxyphenylpyruvate dioxygenase (HPPD) is a crucial enzyme in tyrosine metabolism and a potential target for herbicide and drug discovery. This study summarizes the structures of HPPD complexed with various molecules and analyzes the inhibitory mechanisms of different inhibitors. By identifying key structural factors, the study provides insights into the design of new inhibitors with enhanced potency and selectivity.

Survey on the Recent Advances in 4-Hydroxyphenylpyruvate Dioxygenase (HPPD) Inhibition by Diketone and Triketone Derivatives and Congeneric Compounds: Structural Analysis of HPPD/Inhibitor Complexes and Structure-Activity Relationship Considerations

Paolo Governa et al.

Summary: The serendipitous discovery of HPPD inhibitors from allelopathic plants has paved the way for the search of new and effective herbicidal agents. Various active and selective compounds have been found, including cyclohexane-based triketones, pyrazole-based diketones, and diketonitriles. Efforts have also been made to enhance inhibitory constant and herbicidal activity, as well as to broaden weed control and ensure crop safety. HPPD inhibitors have also shown potential as therapeutic agents and insecticides. Structure-activity relationship analysis can provide suggestions for the development of next generation optimized compounds.

Effects of the novel HPPD-inhibitor herbicide QYM201 on enzyme activity and microorganisms, and its degradation in soil

Fengwen Zhang et al.

Summary: QYM201 initially inhibited soil enzyme activity and microorganism quantities, with effects decreasing over time and eventually returning to control levels. Catalase activity was stimulated by QYM201, while microorganisms responded differently to different concentrations of QYM201. The half-life and degradation rate constant of QYM201 in the studied soils were determined.

Unravelling Phytotoxicity and Mode of Action of Tripyrasulfone, a Novel Herbicide

Hengzhi Wang et al.

Summary: Tripyrasulfone is a new herbicide that acts by blocking the biosynthesis of HGA in plants. It affects pigment composition and photosynthetic activity, leading to leaf necrosis and plant death. Additionally, Tripyrasulfone tend to be hydrolyzed in plants, inhibiting the activity of HPPD enzymes and causing damages to the photosystem II complex.

Rational Redesign of Enzyme via the Combination of Quantum Mechanics/Molecular Mechanics, Molecular Dynamics, and Structural Biology Study

Hong-Yan Lin et al.

Summary: The study introduces a rational redesign strategy for enzyme engineering based on QM/MM calculation and molecular dynamic simulations, leading to the discovery of new HPPD mutants and products. The findings not only enhance understanding of the HPPD catalytic pathway but also demonstrate a generally applicable platform for biocatalyst development.
