Engineering yeast for high-level production of diterpenoid sclareol

Summary: This study used a modular approach to globally rewire the cellular metabolism and achieved a sclareol production of 11.4 g/L, the highest reported titer in microbes. Metabolic flux analysis showed that modular balanced metabolism directed the metabolic flux towards the biosynthesis of targeted molecules, and transcriptomic analysis revealed that the expression of central metabolism genes was shaped for a new balanced metabolism, which laid a foundation for extensive metabolic engineering of other microbial species for sustainable bio-production.

Engineering a powerful green cell factory for robust photoautotrophic diterpenoid production

Summary: In this study, innovative synthetic biology and efficient metabolic engineering strategies were combined to redirect metabolic flux through the MEP pathway in the green microalgae, C. reinhardtii, for efficient heterologous diterpenoid synthesis. Engineering of key enzymes and overexpression of fusion proteins successfully increased the production of high-value diterpenoids. Through fully photoautotrophic high cell density cultivations, significant amounts of sclareol, a high-value diterpenoid, were produced, demonstrating the potential of green microalgae as a powerful phototrophic cell factory for industrial biotechnology.
