Chromosomal editing of Corynebacterium glutamicum ATCC 13032 to produce gamma-aminobutyric acid

Chengzhen Yao et al.

Summary: A high GABA-producing strain of Corynebacterium glutamicum was constructed by chromosomal editing, without the need for antibiotics or inducers. The strategy of enhancing l-glutamic acid synthesis and optimizing the copy numbers of GABA-related genes resulted in increased GABA production.

Engineering of Corynebacterium glutamicum for high-level γ-aminobutyric acid production from glycerol by dynamic metabolic control

Liang Wei et al.

Summary: Synthetic biology aims to reprogram microbial cells for efficient production of value-added compounds from low-cost renewable substrates. One major challenge in chemical biosynthesis is the competition between cell metabolism and target product synthesis for limited cellular resources. In this study, a tunable growth phase-dependent autonomous bifunctional genetic switch (GABS) was created to dynamically redirect carbon flux for metabolic state switching from cell growth mode to production mode, resulting in high-level GABA production from low-value glycerol in Corynebacterium glutamicum.

Adaptive laboratory evolution of Escherichia coli W enhances gamma-aminobutyric acid production using glycerol as the carbon source

Kangsan Kim et al.

Summary: This study utilized evolutionary and metabolic engineering to improve an Escherichia coli strain for more efficient GABA production from glycerol. Through mutation analysis and metabolic pathway regulation, the GABA yield and specific productivity of the glycerol-adapted strain were significantly increased compared to the wild-type.

Effects of Synonymous Mutations beyond Codon Bias: The Evidence for Adaptive Synonymous Substitutions from Microbial Evolution Experiments

Susan F. Bailey et al.

Summary: Synonymous mutations are not always neutral, as they can drive adaptive evolution through impacts on gene expression and protein folding. Site-directed mutagenesis experiments show that the fitness effects of synonymous mutations are more similar to nonsynonymous mutations, especially for beneficial mutations. Experimental evolution studies also demonstrate that synonymous mutations can impact fitness through various mechanisms beyond just being neutral.

Enhancing the production of γ-aminobutyric acid in Escherichia coli BL21 by engineering the enzymes of the regeneration pathway of the coenzyme factor pyridoxal 5′-phosphate

Ping Yu et al.

Summary: The study focused on engineering the enzymes involved in the regeneration pathway of PLP to increase GABA production in E. coli. Initial concentration of L-MSG was found to affect GABA production, with higher concentrations yielding more GABA. Introduction of the regeneration pathway enzymes increased GABA production efficiency in E. coli BL21(DE3).
