Paraquat mediates BV-2 microglia activation by raising intracellular ROS and inhibiting Akt1 phosphorylation

Summary: This study investigated the mechanisms of inflammation induced by paraquat (PQ) in BV-2 microglia cells and the role of the ROS/Akt1 pathway. The results showed that PQ activated BV-2 cells and increased the expression of cell activation markers, leading to an increase in the M1 phenotype. PQ also induced the generation of reactive oxygen species (ROS) and inhibited AKT1 phosphorylation. Treatment with a ROS scavenger decreased the M1 phenotype and restored p-Akt1 levels. Furthermore, overexpression of Akt1(S473E) partially attenuated the PQ-induced increase in M1 phenotype. These findings suggest that PQ activates BV-2 microglia through increased ROS-mediated Akt1 inhibition, leading to neuroinflammation.
