Dealing with Macrophage Plasticity to Address Therapeutic Challenges in Head and Neck Cancers

Sonia Furgiuele et al.

Summary: The head and neck tumor microenvironment is infiltrated with macrophages, particularly tumor-associated macrophages (TAM/M2-like), which are associated with poor overall survival in head and neck cancers. The plasticity of macrophages enables their repolarization towards an anti-tumoral phenotype (M1), making it a promising cancer treatment strategy. This study evaluates macrophage infiltration in HNC patients, optimizes a polarization protocol, and confirms the plasticity of the model using M1 activators.

Immunotherapy for cancer: effects of iron oxide nanoparticles on polarization of tumor-associated macrophages

Camila Sales Nascimento et al.

Summary: Cancer immunotherapy focuses on activating the patient's immune system against cancer, while interest in metabolic reprogramming of tumor-associated macrophages has increased as a therapeutic strategy. Iron metabolism is a key in reprogramming macrophages to a tumor suppressor phenotype, with iron oxide nanoparticles emerging as a potential anti-tumor strategy.

Iron Metabolism in the Tumor Microenvironment-Implications for Anti-Cancer Immune Response

Alessandro Sacco et al.

Summary: New insights have been gained into the impact of iron metabolism within the tumor microenvironment on cancer development. Iron promotes the production of reactive oxygen species, which can trigger cell death or malignant transformation, affecting tumor immunosurveillance. The potential and controversies of using iron metabolism as a target for anticancer treatments are discussed, particularly in the context of hot and cold tumors.

Macrophages and Iron: A Special Relationship

Stefania Recalcati et al.

Summary: Macrophages play a crucial role in the immune response, with a principal function of phagocytic clearance of pathogens, apoptotic, and senescent cells. They also have a unique and important role in iron trafficking, recycling large quantities of iron obtained from hemoglobin degradation and handling toxic levels of iron.

Monocyte Infiltration and Differentiation in 3D Multicellular Spheroid Cancer Models

Natasha Helleberg Madsen et al.

Summary: In this study, a 3D multicellular spheroid model was established to analyze monocyte infiltration and differentiation in the presence of cancer cells and fibroblasts. The differentiation of monocytes into macrophages with diverse phenotypes was found to be influenced by the specific cancer cell line used in the model. Analysis of cytokine secretion in the spheroid cultures revealed a significant difference depending on the type of cancer cell line, with increased production of inflammatory and pro-tumorigenic factors upon the addition of monocytes.
