Liposomes embedded with PEGylated iron oxide nanoparticles enable ferroptosis and combination therapy in cancer

Summary: A novel drug delivery system, Lp-IO, was developed to induce lipid peroxidation and ferroptosis in cancer cells. Lp-IO integrated ultrasmall iron oxide nanoparticles, MRI tracing, and pH/ROS-sensitive drug delivery for combination cancer therapies. It achieved synergistic antineoplastic effects and reduced chemotherapeutic toxicity by delivering doxorubicin.

In vitro model for predicting the access and distribution of drugs in the brain using hCMEC/D3 cells

Summary: This study aimed to improve a previously proposed in vitro cell-based model by using a more physiological cell line and established four linear IVIVCs to predict the pharmacokinetic parameters related to drug access and distribution in the CNS. The results showed that the best IVIVCs were those correlating in vitro and in vivo parameters related to the binding rate of a substance to brain tissue, highlighting the importance of using brain homogenate in in vitro brain permeability assays.

Safety of Gliadel Implant for Malignant Glioma: Report of Postmarketing Surveillance in Japan

Summary: A postmarketing surveillance study in Japan evaluated the safety of Gliadel Wafer in treating malignant glioma patients, with 558 patients enrolled. Most patients received eight Gliadel wafers and the most common adverse reactions included cerebral edema, convulsion, impaired healing, and infection. The study suggests that the risk of toxicity with Gliadel placement is relatively tolerable, but clinicians should carefully evaluate the survival benefits and possible toxicities before using Gliadel.
