Enhanced osteogenic effect in reduced BMP-2 doses with siNoggin transfected pre-osteoblasts in 3D silk scaffolds

Summary: Amyotrophic lateral sclerosis (ALS), a neurodegenerative disease with a very poor prognosis, is hindered by the blood-brain barrier (BBB), which restricts the access of drugs commonly used in ALS to the brain. In this study, riluzole-loaded nanostructured lipid carriers (NLC) functionalized with lactoferrin (Lf) were prepared to overcome this limitation and increase brain targeting. The results showed that Lf-functionalized NLC exhibited a promising ability to target riluzole to the brain without causing significant toxicity to cells.

Injecting a fibrin gel with sulfated chitosan nanoparticles can improve bone regeneration and reduce inflammation at the fracture site. Summary: The study focuses on developing polymeric nano-particles as a local drug delivery system for promoting alveolar bone regeneration. Different parameters such as PVA concentration, drug/polymer ratio, inner phase content, polymer type, and pH of the external water phase were investigated for their effects on the characteristics of nanoparticles. Encapsulation efficiency highly depends on PVA concentration and pH, while the mean particle size decreases with the decrease in PLGA molecular weight. Sustained release for up to 3 months was achieved for all formulations using clindamycin phosphate.

To address this issue, researchers developed a novel bioink by incorporating bone morphogenetic protein-2 (BMP-2)-loaded nanoparticles into alginate, which improved the mechanical properties and printability of the bioink. In vitro studies showed that this bioink significantly induced osteogenesis of mesenchymal stem cells (MSCs).
