Mitochondrial STAT3 exacerbates LPS-induced sepsis by driving CPT1a-mediated fatty acid oxidation

Rongqing Li et al.

Summary: The study revealed that mitochondrial STAT3 in macrophages induced NF-kappa B nuclear localization and exacerbated LPS-induced sepsis, along with promoting a shift from glucose metabolism to fatty acid oxidation. Furthermore, mitochondrial STAT3 inhibited the ubiquitination and degradation of carnitine palmitoyl transferase 1a (CPT1a) in LPS-treated macrophages, with an interaction observed between CPT1a and ubiquitin-specific peptidase 50 (USP50).

Uridine alleviates LPS-induced ARDS and improves insulin sensitivity by decreasing oxidative stress and inflammatory processes

Lei Zhang et al.

Summary: This study investigated the effect of uridine on acute respiratory distress syndrome (ARDS) and insulin resistance. The results showed that uridine treatment significantly alleviated lung tissue damage and down-regulated the levels of proinflammatory cytokines. Uridine also decreased the numbers of total cells and neutrophils in the bronchoalveolar lavage fluid. The study further revealed that uridine treatment down-regulated NF-kappa B signaling and enhanced insulin sensitivity in the ARDS mice model. Furthermore, uridine treatment reduced inflammation and oxidative stress levels. These findings suggest that uridine may serve as a potential drug for the treatment of ARDS and insulin resistance.

Guanosine and uridine alleviate airway inflammation via inhibition of the MAPK and NF-κB signals in OVA-induced asthmatic mice

Yujiao Luo et al.

Summary: The study investigated the anti-inflammatory effects of guanosine and uridine in an asthma model, showing that both nucleosides can reduce lung inflammation by suppressing the MAPK and NF-kappa B pathways. Treatment with guanosine or uridine decreased total cell counts, cytokines levels, OVA-specific IgE, and mRNA expression in asthmatic mice. Additionally, the production of IL-6 in THP-1 cells and the expression of key molecules in the MAPK and NF-kappa B pathways were reduced by both nucleosides, indicating their potential therapeutic value in asthma.
