Formation of mucus-permeable nanoparticles from soy protein isolate by partial enzymatic hydrolysis coupled with thermal and pH-shifting treatment

Summary: Modulating the size and surface charge of nanocarriers can enhance the bioavailability of encapsulated cargos in mucus by increasing mucus penetration. In this study, mucus-permeable soy protein nanoparticles (SPNPs) were fabricated using gastrointestinal proteolysis, heating, and pH-shifting treatment. The results showed that different treatments led to different subunit assemblies and structures, resulting in different mucus permeabilities. This study provides an alternative method to fabricate mucus-permeable nanoparticles and establishes a relationship between protein subunit assembly behavior and mucus permeability.

It was found that the secondary masking of intermediate polypeptides restricted the opening of the substrate. Additionally, demasking and secondary masking were shown to compete with each other.

Summary: High-resolution ultrasonic spectroscopy was used for real-time monitoring of beta-casein hydrolysis by trypsin for the first time, providing kinetic and mechanistic description without requiring optical transparency. The exponential dependence of kinetic parameters allowed determination of activation and deactivation energies of hydrolysis. The ultrasonic assay allowed rapid detection of trypsin activity even at sub-nanomolar concentration.

Atomic force microscopy characterization of β-casein nanoparticles on mica and graphite

Summary: Using atomic force microscopy, it was found that the morphology and size distribution of beta-casein nanoparticles adsorbed on mica and graphite were different. The height of dried nanoparticles on graphite was approximately half that on mica, and the cross-sectional profiles of the particles varied between these two substrates, attributed to the hydrophobic interaction between nanoparticles and the graphite surface.
