Current State and Future Directions of EGFR-Directed Therapy in Head and Neck Cancer

Praveena Tathineni et al.

Summary: EGFR is often overexpressed in HNSCC, but the clinical outcomes of EGFR-directed therapy have not been significant. Cisplatin is superior to cetuximab in curative-intent treatment, while carboplatin-based treatment is preferred for cisplatin-ineligible patients. EGFR inhibitors have a stronger role in recurrent/metastatic HNSCC, particularly for patients who have progressed on immune checkpoint inhibitors. Overall, EGFR-directed therapies have shown modest value, and the future may lie in combination therapies with immunotherapy.

Wnt Signaling Inhibitors and Their Promising Role in Tumor Treatment

Nives Pecina-Slaus et al.

Summary: In the search for improved antitumor therapies, inhibition of the Wnt signaling pathway has emerged as a promising target due to its altered functioning in human tumors. The pathway's frequent mutations and epigenetic changes in tumors have promoted the investigation of Wnt signaling inhibitors. Studies have shown that knocking down specific components of the pathway inhibits tumor growth. This review discusses the inhibitors of the canonical Wnt pathway, evaluates their effectiveness as therapeutics, and examines their potential side effects.

Therapeutic Potential of Naturally Occurring Small Molecules to Target the Wnt/beta-Catenin Signaling Pathway in Colorectal Cancer

Luiz F. S. Oliveira et al.

Summary: Colorectal cancer (CRC) is the second leading cause of death worldwide, and its treatment is limited due to late diagnoses and resistance to available treatments. The use of naturally occurring small molecules that inhibit the Wnt/beta-catenin pathway in experimental CRC models has shown promise in overcoming these challenges. However, moving these small molecules from the laboratory to the clinical platform presents its own set of progress and challenges. This review discusses the importance of understanding the role of the Wnt/beta-catenin pathway in CRC and highlights the potential of naturally occurring small molecules as therapeutic agents.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.
