The oncogenic driving force of CD30 signaling-induced chromosomal instability in adult T-cell leukemia/lymphoma

Makoto Nakashima et al.

Summary: Adult T-cell leukemia/lymphoma (ATL) develops through the accumulation of gene mutations and chromosome aberrations. CD30 signaling induces chromosomal instability and clonal expansion in ATL cells. CD30 expression is correlated with genomic instability, suggesting its role as an oncogenic factor in ATL progression with clonal evolution.

Summary: The study developed a capture-based sequencing panel and identified distinct genomic and transcriptomic features associated with subclonal evolution in Adult T-cell leukemia-lymphoma (ATL). Subclonal genetic heterogeneity and diverse gene expression present serious challenges in understanding cancer behavior and therapeutic strategies.

A novel model of alternative NF-κB pathway activation in anaplastic large cell lymphoma

Hongbo Wang et al.

Summary: Aberrant activation of NF-kappa B is a key oncogenic mechanism in B-cell lymphoma, while its role in anaplastic large cell lymphomas (ALCL) remains unclear. Study on ALCL cell line models revealed supporting roles of NFKB2 and NIK pathway, with STAT3 identified as the major mediator for NIK-dependent NF-kappa B activation in ALCL. The findings suggest potential benefits of combined NIK and JAK inhibitor therapy for NIK-positive ALK- ALCL with JAK/STAT3 somatic mutations.
