Aberrant Expression and Prognostic Potential of IL-37 in Human Lung Adenocarcinoma

Summary: This study explores the prognostic and diagnostic potential of Interleukin-37 (IL-37) in human tumors, particularly in lung adenocarcinoma (LUAD). The results show that IL-37 is associated with LUAD development and tumor stage, and high mRNA levels of IL-37 are favorable prognostic factors for patients' overall survival. Additionally, IL-37 is linked with distinct infiltration patterns of certain cell subsets that control LUAD anti-tumor immune responses. These findings suggest that IL-37 and its receptor SIGIRR could serve as biomarkers and immune-checkpoint therapeutic targets for LUAD patients.

ARID1A-deficient bladder cancer is dependent on PI3K signaling and sensitive to EZH2 and PI3K inhibitors

Summary: The study reveals that bladder cancers with ARID1A mutations are more sensitive to EZH2 inhibitors and/or PI3K inhibitors. Mechanistic studies demonstrate that ARID1A deficiency leads to a dependency on PI3K signaling, and EZH2 inhibitor sensitivity is associated with upregulation of PIK3IP1. Moreover, combination therapy with EZH2 and PI3K inhibitors shows a synergistic effect on ARID1A-deficient bladder cancer.

Interleukin-37 promotes colitis-associated carcinogenesis via SIGIRR-mediated cytotoxic T cells dysfunction

Summary: Interleukin-37b (IL-37) is a crucial inhibitor of natural and acquired immunity. Research has shown that IL-37 increases the risk of colitis-associated colorectal cancer (CAC) and dampens the immune response mediated by cytotoxic T cells. IL-37 has little effect on intestinal mutagenesis, CRC cell proliferation, apoptosis, and migration. Additionally, IL-37 levels are significantly increased in CRC patients, positively correlated with serum CRC biomarker CEA levels, but negatively correlated with CD8(+) T cell infiltration.
