Platelet-derived microparticles stimulated by anti-β2GPI/β2GPI complexes induce pyroptosis of endothelial cells in antiphospholipid syndrome

Summary: This study investigated the specific mechanism of endothelial cell pyroptosis induced by platelet-released PMPs activated by anti-beta(2)GPI/beta(2)GPI complexes. The increased expression of NLRP3 in PMPs stimulated by anti-beta(2)GPI/beta(2)GPI complexes is closely associated with GSDMD-N, a protein involved in endothelial pyroptosis. The findings provide new insights into the mechanism of endothelial cell damage in APS and identify a potential drug target for the clinical treatment of APS.

Summary: Citrullinated neoepitopes have been identified as key triggers for the synthesis of anti-citrullinated protein antibodies (ACPA) in rheumatoid arthritis (RA) patients. This study investigated the expression of citrullinated proteins and enzymes in human platelets and platelet-derived microparticles (PDP). The results revealed the presence of citrullinated autoantigens in platelets and PDPs, suggesting that they could be potential targets for ACPA in RA. These findings provide valuable insights into the role of platelets in triggering ACPA production in RA.

This imbalance in platelet clearance may contribute to vascular inflammation and fibrosis in SSc.

These diseases have a lengthy prodromal phase with a gradually evolving interaction between genetic profile and environment.

Microbiota dysbiosis in primary Sjogren's syndrome and the ameliorative effect of hydroxychloroquine

Summary: The human microbiome plays a significant role in the pathogenesis of primary Sjogren's syndrome (pSS). Dysbiosis of the microbiota in the gut, oral cavity, and vagina is evident in pSS patients, with the oral microbiome showing the most significant variation. Key indicator bacteria and clinical characteristics are identified in different body sites, suggesting the importance of microbial dysbiosis in pSS. Furthermore, similar dysbiosis is observed in individuals with pre-clinical pSS or non-pSS-related diseases, indicating that microbial shifts may occur prior to the development of pSS. Hydroxychloroquine (HCQ) treatment partially resolves microbial dysbiosis in pSS patients, although the microbiota composition remains disordered.
